Objectives: The incidence of acute kidney injury (AKI) in infective endocarditis (IE), its risk factors and consequences on patient and renal survival remain debated. Methods: Patients hospitalized for a first episode of IE (possible or definite according to modified Duke criteria) between 2013 and 2016 were included. The primary endpoint was to determine risk factors for early AKI (E-AKI) during the first week of management of IE. Results: A total of 276 patients were included: 220 (79.7%) had definite IE and 56 (20.3%) had possible IE. E-AKI occurred in 150 patients (53%). IE due to Staphylococcus aureus (OR 3.41; 95% CI 1.83-6.39; p < 0.01), history of diabetes (OR 2.34; 95% CI 1.25-4.37; p < 0.01), peripheral arterial disease (OR 2.59; 95% CI 1.07-6.23; p < 0.05), immunological manifestations (OR 3.11; 95% CI 1.31-7.39; p = 0.01), and use of norepinephrine (OR 3.44; 95% CI 1.72-7.02; p < 0.01) were associated with E-AKI. In subgroup analysis, infectious disease consultation was associated with a lower risk of AKI at day 7 (OR 0.41; 95% CI 0.16-0.88; p = 0.04). E-AKI was associated with 1-year mortality (OR 1.65; 95% CI 1.03-2.64; p = 0.04) and chronic kidney disease progression (OR 2.23; 95% CI 1.30-3.82; p < 0.01). Conclusions: E-AKI is common in IE and often associated with non-modifiable variables. Multidisciplinary management should be mandatory, and awareness of AKI diagnosis and etiological explorations should be raised. (C) 2020 The Authors. Alpinia galanga is a wild medicinal plant having tremendous industrial importance. However, lack of quality planting material in natural resources restricted its wide exposure in industrial sector. Thus, a standardised protocol for stable supply of quality planting materials with genetical and phytochemical stability is need of the hour to meet commercial requirements. The current study reports an efficient protocol to regenerate micropropagated plantlets with long term stability in molecular as well as phytochemical level. Murashige and Skoog (MS) media augmented with 3 mg/l 6-Benzyladenine (BA), 3 mg/l Kinetin (Kn) and 1 mg/l Napthalene Acetic acid (NAA) was found optimum for regeneration and multiplication of plantlets. In order to establish genetic stability molecular marker-based profiling of micropropagated plants were done at every 6 months intervals up to 6 years. Monomorphic banding pattern were found that are identical to the mother plants. After six years of in vitro conservation, the regenerated plants were transplanted to the field and its drug yielding potential were evaluated through phytoconstituent analysis along with bioactivity studies. A comparative Gas chromatography (GC) and mass spectroscopy (MS) analysis of essential oils showed no significant differences in phytoconstituents between the conventional and in vitro propagated plants. The bioactivities like antioxidant, antimicrobial and anticancerous study along with total phenolic and total flavonoid content of in vitro plants were compared with mother plants and found stable with negligible variation. Thus, the present study has enough significance towards commercial exploitation of A. galanga with phytochemical and molecular stability.

Vicinal dibromides and dichlorides are important commodity chemicals and indispensable synthetic intermediates in modern chemistry that are traditionally synthesized using hazardous elemental chlorine and bromine. Meanwhile, the environmental persistence of halogenated pollutants necessitates improved approaches to accelerate their remediation. Here, we introduce an electrochemically assisted shuttle (e-shuttle) paradigm for the facile and scalable interconversion of alkenes and vicinal dihalides, a class of reactions that can be used both to synthesize useful dihalogenated molecules from simple alkenes and to recycle waste material through retro-dihalogenation. The reaction is demonstrated using 1,2-dibromoethane, as well as 1,1,1,2-tetrachloroethane or 1,2-dichloroethane, to dibrominate or dichlorinate, respectively, a wide range of alkenes in a simple setup with inexpensive graphite electrodes. Conversely, the hexachlorinated persistent pollutant lindane could be fully dechlorinated to benzene in soil samples using simple alkene acceptors.

Artemisia fragrans is an aromatic and perennial herb with green silver leaves and yellow flowers, widely distributed in Iran, Azerbaijan, Russia and Turkey. The plant and its essential oil (EO) are used in traditional medicine and have economic value. In this study, the composition, antibacterial and antioxidant activities of 16 A. fragrans essential oil samples collected from different habitats in Iran were analyzed. A total of 85 compounds were detected using GC-MS analysis, the major constituents being camphor (9.91-34.44 %), alpha-thujone (19.22-42.63 %) and 1,8-cineole (12.57-31.87 %). Davanone D, alpha-cadinol, verbenene, ortho-oci-men were detected in the analysis as the new components for this species and are first reported by us. Anti-bacterial activity and minimum inhibitory and bactericidal concentrations of EOs were tested on Escherichia coli, Klebsiella pneumonia and Proteus vulgaris (Gram-negative strains) Staphylococcus aureus, Staphylococ-cus epidermidis and Bacillus subtilis (Gram-positive strains). The EOs showed better inhibitory effects against Gram-negative strains compared to Gram-positive ones. Among the EOs, E7 (IC50 = 9.1 mu g/mL) population showed the highest radical scavenging capacity, which was equal to the positive control (butylated hyroxytoluene). The quantitative and qualitative variability of the studied EOs and their antibacterial and antioxidant activities may be recommended for pharmacological and cosmetic industries.

Buckwheat was processed by solid-state fermentation (SSF) with the probiotic fungal strain Eurotium cristatum YL-1. The effects of SSF on the phytochemical content, as well as the antioxidant and alpha-glucosidase inhibitory activities, on buckwheat were revealed. Metabolite differences between non-fermented buckwheat (BW) and E. cristatum fermented buckwheat (FBW) were investigated by LC-MS/MS-based untargeted metabolomics. Results showed that 103 and 68 metabolites remarkably differed between BW and FBW in positive and negative ionization modes, respectively. Most phenolic compounds and alkaloids were significantly up-regulated during SSF. Hydrolytic enzymes (i.e., beta-glucosidase, alpha-amylase, protease, and cellulase) were produced by the filamentous fungus E. cristatum during SSF. In vitro spectrophotometric assays demonstrated that the total phenolics content, ferric reducing antioxidant power, reducing power, scavenging activities of DPPH radical and ABTS center dot+, and alpha-glucosidase inhibitory activity of buckwheat were considerably enhanced after processing by SSF with E. cristatum. Additionally, solvents with different polarities significantly influenced the antioxidant and alpha-glucosidase inhibitory activities of buckwheat extracts. Our study indicated that processing by SSF with E. cristatum can greatly improve the phytochemical components of buckwheat and consequently contribute to its antioxidant and alpha-glucosidase inhibitory activities. SSF with E. cristatum is an innovative method for enhancing the health-promoting components and bioactivities of buckwheat.

Styrene can be formed by the microbial metabolism of bacteria and fungi. In our previous study, styrene was determined as a spoilage marker of Fuji apples decayed by Penicillium expansum, which is responsible for postharvest diseases. In the present study, P. expansum was cultivated in potato dextrose broth added with phenylalanine-which is a precursor of styrene-using different initial pH values and cultivation times. Volatile compounds were extracted and analyzed using gas chromatography-mass spectrometry (GC-MS) combined with stir-bar sorptive extraction. The 76 detected volatile compounds included 3-methylbutan-1-ol, 3-methyl butanal, oct-1-en-3-ol, geosmin, nonanal, hexanal, and gamma-decalactone. In particular, the formation of 10 volatile compounds derived from phenylalanine (including styrene and 2-phenylethanol) showed different patterns according to pH and the cultivation time. Partial least square-discriminant analysis (PLS-DA) plots indicated that the volatile compounds were affected more by pH than by the cultivation time. These results indicated that an acidic pH enhances the formation of styrene and that pH could be a critical factor in the production of styrene by P. expansum. This is the first study to analyze volatile compounds produced by P. expansum according to pH and cultivation time and to determine their effects on the formation of styrene.

The floral scents of three forms of cultivated Plumeria rubra L. were evaluated through mass flowering phenology using the dynamic headspace adsorption method and were identified with coupled gas chromatography and mass spectrometry. The forms P. rubra f. acutifolia and P. rubra f. lutea had white and yellow flower petals, respectively, and the flower petals of P. rubra f. rubra were red. Although 68 components of the flower scents of the three forms were recorded in different proportions, only 14 chemical compounds were identified with statistically significance. The main volatile compounds in the red form of P. rubra L. were fatty acid derivatives (56.75%). The main compounds in the white and yellow forms of P. rubra L. were benzenoid and terpene, with proportions of 48.38% and 33.33% in P. rubra f. acutifolia and proportions of 42.30% and 47.43% in P. rubra f. lutea, respectively. These differences in the flower scents might be one result of the minor genetic differences between these forms, similar to the role of genetic differences in the flower color combinations of the three forms. We conclude that petal color traits can, to some extent, reflect differences in floral scent compositions and that minor genetic differences of different plant forms exert impacts not only on flower color but also on the phytochemistry of floral scents.

Although the evolutionary significance of the early-diverging flowering plant Amborella (Amborella trichopoda Baill.) is widely recognized, its metabolic landscape, particularly specialized metabolites, is currently underexplored. In this work, we analyzed the metabolomes of Amborella tissues using liquid chromatography high-resolution electrospray ionization mass spectrometry (LC-HR-ESI-MS). By matching the mass spectra of Amborella metabolites with those of authentic phytochemical standards in the publicly accessible libraries, 63, 39, and 21 compounds were tentatively identified in leaves, stems, and roots, respectively. Free amino acids, organic acids, simple sugars, cofactors, as well as abundant glycosylated and/or methylated phenolic specialized metabolites were observed in Amborella leaves. Diverse metabolites were also detected in stems and roots, including those that were not identified in leaves. To understand the biosynthesis of specialized metabolites with glycosyl and methyl modifications, families of small molecule UDP-dependent glycosyltransferases (UGTs) and O-methyltransferases (OMTs) were identified in the Amborella genome and the InterPro database based on conserved functional domains. Of the 17 phylogenetic groups of plant UGTs (A-Q) defined to date, Amborella UGTs are absent from groups B, N, and P, but they are highly abundant in group L. Among the 25 Amborella OMTs, 7 cluster with caffeoyl-coenzyme A (CCoA) OMTs involved in lignin and phenolic metabolism, whereas 18 form a clade with plant OMTs that methylate hydroxycinnamic acids, flavonoids, or alkaloids. Overall, this first report of metabolomes and candidate metabolic genes in Amborella provides a starting point to a better understanding of specialized metabolites and biosynthetic enzymes in this basal lineage of flowering plants.

The fumigant antibacterial activities of 50 plant essential oils belonging to 10 families were investigated against Agrobacterium tumefaciens. Among the test plant essential oils, Cinnamomum verum (cinnamon bark) essential oil showed the most potent fumigant antibacterial activity. When we investigated the antibacterial activities of compounds identified from cinnamon bark essential oil and 9 congeners of trans-cinnamaldehyde, lengths of inhibition zone of trans-cinnamaldehyde, salicylaldehyde and hydrocinnamaldehyde were 1.28, 1.73, and 1.24 cm at 0.625 mg/paper disc concentration, respectively. To determine the mode of action of trans-cinnamaldehyde and salicylaldehyde, intercellular reactive oxygen species (ROS) generation and cell membrane integrity were determined using a confocal laser scanning microscopy. Furthermore, we compared the up- and down-regulated gene expression of A. tumefaciens treated with trans-cinnamaldehyde and salicylaldehyde with that of untreated A. tumefaciens. With cutoffs of vertical bar log2FC vertical bar > 1 and FDR < 0.05, 29 and 43 down-regulated genes and 27 and 117 up-regulated genes were found in the treatment of trans-cinnamaldehyde and salicylaldehyde, respectively. Based on the ROS generation results, cell membrane integrity assay, and gene expression, we conclude that the antibacterial mode of action of trans-cinnamaldehyde and salicylaldehyde is ROS generation by the Fenton reaction caused by the down-regulation of an ATP synthesis-related gene cluster, corrupted iron ion homeostasis, and a corrupted ROS defense mechanism. The high concentration of ROS damaged the A. tumefaciens cell membrane, which caused cell death.
