Left ventricular hypertrophy (LVH) is an important characteristic of hypertensive heart disease. Renin-angiotensin system (RAS) blockers have been shown to be effective drugs for the reversal of LVH. Clinical and experimental studies have shown that Zi Shen Huo Luo Formula (ZSHLF) can improve the efficacy of perindopril in the treatment of hypertensive LVH, but its mechanism is unclear. This study aimed to investigate the possible mechanism to improve the efficacy of perindopril. First, we identified 23 compounds in ZSHLF by ultra performance liquid chromatography/tandem mass spectrometry (UPLC-MS/MS) analysis, among which ferulic acid, caffeic acid, vanillic acid, berberine, rutin, quercetin, kaempferol, stachydrine, and tiliroside have been reported to reduce blood pressure and exhibit cardioprotective effects. Second, we treated spontaneously hypertensive rats (SHRs) with perindopril and ZSHLF for 12 continuous weeks and found that chronic use of perindopril could increase the aldosterone (ALD) levels and cause aldosterone breakthrough (ABT). ZSHLF combined with perindopril reduced the ALD levels, interfered with ABT, decreased blood pressure, improved left ventricular diastolic dysfunction, and decreased the collagen volume fraction; these effects were superior to those of perindopril alone. In vitro experiments, ALD-induced cardiomyocytes (H9c2 cells) and cardiac fibroblasts were treated with ZSHLF-containing serum, which suppressed ALD-induced cardiomyocyte hypertrophy and cardiac fibroblast proliferation, increased mineralocorticoid receptor (MR) and Cav-1 colocalization and decreased phosphorylated epidermal growth factor receptor (pEGFR) and phosphorylated extracellular signal-regulated kinase (pERK) protein expression the cells. In conclusion, ZSHLF can interfere with ABT and affect the pathological role of ALD by affecting MR and Cav-1 interactions and EGFR/ERK signaling pathway. These effects represent a possible mechanism by which ZSHLF improves the efficacy of angiotensin-converting enzyme inhibitors (ACEIs) in hypertensive LVH treatment. However, the major bioactive components or metabolites responsible for the effects and the implications of these findings in patients need further verification.

A Ni salt of phosphomolybdic acid (NiHPMA) was synthesized and characterized by various physico-chemical techniques such as EDX, UV-Visible spectroscopy, FT-IR, Raman spectroscopy and XPS. FT-IR and Raman spectroscopy confirm the presence of Ni as a counter cation while UV-Visible and XPS studies to confirm the presence of Ni(ii) in the catalyst. The catalyst was evaluated for its bi-functional activity towards the tandem conversion of benzaldehyde to ethyl benzoate and it was found that very small amounts of Ni (2.64 x 10(-3)mmol) enhance the selectivity towards benzoate. A detailed mechanistic study was carried out by UV-Visible and Raman spectroscopy to confirm that both intermediate species, Mo-peroxo and Ni-oxo, are responsible for higher selectivity towards esters. Further, a study to determine the effect of addenda atoms (heteropoly acid) was also carried out. The catalyst was also found to be viable for a number of aldehydes under optimized conditions.

Herein, we report a graphene oxide (GO) catalyzed condensation of non-activated esters and amines, that can enable diverse amides to be synthesized from abundant ethyl esters forming only volatile alcohol as a by-product. GO accelerates ester to amide conversion in the absence of any additives, unlike other catalysts. A wide range of ester and amine substrates are screened to yield the respective amides in good to excellent yields. The improved catalytic activity can be ascribed to the oxygenated functionalities present on the graphene oxide surface which forms H-bonding with the reactants accelerating the reaction. Improved yields and a wide range of functional group tolerance are some of the important features of the developed protocol.

In the present study, the effect of enzyme pretreatment on essential oil recoveries from sweet basil (Ocimum basilicum L.) leaves was evaluated. Moreover, the consideration on the use of hydrodistilled residue by-products as a source of bioactive phytochemicals with antioxidant, antimicrobial, and repellent effects against the stored-grain pest Tribolium castaneum was examined. Results showed that the enzymatic pretreatment increased the extraction yield of essential oil by 400, 417, and 478% in hemicellulase-, cellulase-, and viscozyme-treated samples, respectively. Phenylpropanoids including methyl cinnamate, methyl eugenol, eugenol and estragol were found as the main components, and were particularly abundant in cellulase-treated samples. From the hydrodistilled residue of enzyme-treated samples, better recoveries of total phenols (TPC) (258.3-470.9 mg GAE/g extract) and flavonoids (TFC) (59.4-94.3 mg QE/g extract) were observed. Using the DPPH, ABTS, and FRAP assays, a strong antioxidant activity of the rosmarinic-rich extract was observed. Such an activity which was mediated through electron transfer mechanism was highly correlated with the TPC, TFC and rosmarinic acid content. The in vitro bioassay showed that methanol extract (6.29 and 12.58 mu L/cm(2)) had repellent activity against the stored-grain pest Tribolium castaneum. These results suggest the potential of enzyme pretreatment to promote the use of hydrodistilled residue by-products as a valuable source of natural antioxidants and repellents ingredients. IRMOF-3 with a high surface area prepared by a hydrothermal method was used for deposition of Pd(OAc)(2) on IRMOF-3 particles. The final catalyst was characterized with FT-IR, nitrogen physisorption, thermogravimetry, scanning electron microscopy, transmission electron microscopy combined with energy dispersive X-ray analysis, wide angle X-ray diffraction spectroscopy and X-ray photoelectron spectroscopy. The reaction parameters such as the type of base, amounts of catalyst and solvents, temperature were optimized. The paper describes synthesis of a novel mesoporous and magnetically regenerated heterogeneous nanocatalyst with high catalytic activity. The synthesis strategy includes a one-step polycondensation reaction of melamine, formaldehyde and FeCl3 center dot 6H(2)O in a simple and sustainable manner and followed by immobilization of Pd(II) on it. Formation and properties of nanocomposite were characterized by different techniques; the results were illustrated good surface area and a well-defined mesopore structure. Also the palladium-catalyzed C-C bond formation between aryl halides and olefins was selected to survey the activity of the synthesized nanocatalyst. Central Composite Design (CCD) strategy was used to predict the optimum synthesis conditions and identify the remarkable and effective factors including temperature, period of the reaction and amount of catalyst. These results were also verified experimentally. In order to minimize systematic error, the experimental runs as a randomized fashion were accomplished.

In this research, the antibacterial activity and the interaction mechanisms of the major chemical constituent present in the Alpinia galanga rhizomes essential oil (AGREO) were investigated. Ethyl cinnamate, methyl cinnamate and n-pentadecane were the major components of AGREO. The antibacterial mechanism of AGREO on Enterohemorrhagic Escherichia coli O157:H7 (EHEC O157) was evaluated and the results proved that the potent antibacterial activity of AGREO was due to higher passive permeability of bacterial cell membrane, followed by crucial intracellular components efflux. Additionally, AGREO disrupted the intracellular physiological metabolism of EHEC O157, including inhibition of P-type ATPases activity and down-regulation expression of four virulence genes associated with EHEC infections. Notably, molecular docking studies suggested that ethyl cinnamate probably competed with the structural NAD(P)(+) for the binding sites of glucose-6-phosphate dehydrogenase (G6PD), resulting in a decrease in G6PD activity with subsequent suppression of respiratory metabolism. The spectroscopic techniques and molecular docking studies proved that the AGREO major components were prone to bind to the minor groove of DNA.

Useful alpha-boryl esters can be synthesized in one step from alpha,beta-unsaturated esters using just a simple to access NHC-BH3 (NHC = N-heterocyclic carbene) and catalytic I-2. The scope of this reductive alpha-borylation methodology is excellent and includes a range of alkyl, aryl substituted and cyclic and acyclic alpha,beta-unsaturated esters. Mechanistic studies involving reductive borylation of a cyclic alpha,beta-unsaturated ester with NHC-BD3/I-2 indicated that concerted hydroboration of the alkene moiety in the alpha,beta-unsaturated ester proceeds instead of a stepwise process involving initial 1,4-hydroboration; this is in contrast to the recently reported reductive alpha-silylation. The BH2(NHC) unit can be transformed into electrophilic BX2(NHC) moieties (X = halide) and the ester moiety can be reduced to the alcohol with the borane unit remaining intact to form b-boryl alcohols. The use of a chiral auxiliary, 8-phenylmenthyl ester, also enables effective stereo-control of the newly formed C-B bond. Combined two step ester reduction/borane oxidation forms diols, including excellent e. e. (97%) for the formation of S-3-phenylpropane-1,2-diol. This work represents a simple transition metal free route to form bench stable alpha-boryl esters from inexpensive starting materials.

Herein we report the first example of an on-water enantiospecific synthesis of alkyl bromides. This procedure allowed the conversion of secondary activated alkyl sulphides to benzylic alkyl bromides, which were obtained in 80-99% yields. The reaction carried out on enantio-pure sulphides provided the corresponding bromides in high yields and enantioselectivity (up to 92% ee; 94% es) at room temperature. The on-water conditions reduced significantly the reaction times compared to similar procedures run in organic media. The condition identified made use of no solvent, required no temperature control and produced a smooth organic phase easily separated for further synthetic use on a multigram-scale without the need for any organic extraction. Therefore, the present constitutes the most operationally simple and environmentally benign approach to a class of much sought organic intermediates.
