Alpinia galanga is a wild medicinal plant having tremendous industrial importance. However, lack of quality planting material in natural resources restricted its wide exposure in industrial sector. Thus, a standardised protocol for stable supply of quality planting materials with genetical and phytochemical stability is need of the hour to meet commercial requirements. The current study reports an efficient protocol to regenerate micropropagated plantlets with long term stability in molecular as well as phytochemical level. Murashige and Skoog (MS) media augmented with 3 mg/l 6-Benzyladenine (BA), 3 mg/l Kinetin (Kn) and 1 mg/l Napthalene Acetic acid (NAA) was found optimum for regeneration and multiplication of plantlets. In order to establish genetic stability molecular marker-based profiling of micropropagated plants were done at every 6 months intervals up to 6 years. Monomorphic banding pattern were found that are identical to the mother plants. After six years of in vitro conservation, the regenerated plants were transplanted to the field and its drug yielding potential were evaluated through phytoconstituent analysis along with bioactivity studies. A comparative Gas chromatography (GC) and mass spectroscopy (MS) analysis of essential oils showed no significant differences in phytoconstituents between the conventional and in vitro propagated plants. The bioactivities like antioxidant, antimicrobial and anticancerous study along with total phenolic and total flavonoid content of in vitro plants were compared with mother plants and found stable with negligible variation. Thus, the present study has enough significance towards commercial exploitation of A. galanga with phytochemical and molecular stability.

Tea is an important beverage source of dietary polyphenols and well known for containing phenolic structure diversity. A series of phenylpropanoid-substituted catechins, flavonols, flavan-3-hexoside, and proanthocyanidin are present in different herbs with various biological activities, inspiring our exploration of phenylpropanoid-substituted ester type of catechins (PSECs) due to the enrichment of galloylated catechins in tea. In this study, we used a guiding-screening-location-isolation integrated route including creating a hypothesized PSEC dataset, MS/MS data acquiring, construction of molecular networks, and traditional column chromatography and preliminarily identified 14 PSECs by MS/MS spectrum. Two of these PSECs were further purified and elucidated by NMR and CD spectra. Further MS detection in tea products and fresh leaves suggests that the production of the two new compounds was enhanced during tea processing. The synthesis mechanism was proposed to obtain these types of components for further investigation on their roles in human health protection. This study provides an example for the exploration of new functional ingredients from food sources guided by MS/MS data-based networking, and also new insights into the reaction mechanism to form new catechin conjugates among polyphenols in green tea.

Green-ripe pineapples are shipped overseas by sea freight, while those picked at full maturity need to be transported by airfreight over the same large distance. In this study, fresh-cut pineapple cubes were assessed two, five, and eight days after processing from green-ripe pineapples after mimicked sea freigh (SF) and fully ripe air-freighted (AF) pineapples. The sea-freighted samples displayed elevated titratable acidity (TA), thus resulting in smaller ratios of total soluble solids and TA compared to the AF pineapples. Differences in the carotenoid levels of the two fresh-cut categories were found to be insignificant. By contrast, hierarchical cluster analysis (HCA) and principal component analysis (PCA) calculated on the basis of the volatiles analysed by headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) permitted to distinguish all six individual sample types and to segregate them into two major clusters (SF and AF). The effect of storage on the volatiles was further evaluated by partial least squares (PLS) regression. Substantial chemical markers to differentiate the individual samples and to describe the effect of storage were deduced from the PCA and PLS regression, respectively. In general, fresh-cut products obtained from fully ripe AF fruit displayed higher concentrations of volatiles, in particular, increased concentrations of diverse methyl esters. With progressing storage duration, the concentrations of ethanol and diverse ethyl esters increased. Moreover, products from AF pineapples displayed lower microbial counts compared to those from SF fruit.

Buckwheat was processed by solid-state fermentation (SSF) with the probiotic fungal strain Eurotium cristatum YL-1. The effects of SSF on the phytochemical content, as well as the antioxidant and alpha-glucosidase inhibitory activities, on buckwheat were revealed. Metabolite differences between non-fermented buckwheat (BW) and E. cristatum fermented buckwheat (FBW) were investigated by LC-MS/MS-based untargeted metabolomics. Results showed that 103 and 68 metabolites remarkably differed between BW and FBW in positive and negative ionization modes, respectively. Most phenolic compounds and alkaloids were significantly up-regulated during SSF. Hydrolytic enzymes (i.e., beta-glucosidase, alpha-amylase, protease, and cellulase) were produced by the filamentous fungus E. cristatum during SSF. In vitro spectrophotometric assays demonstrated that the total phenolics content, ferric reducing antioxidant power, reducing power, scavenging activities of DPPH radical and ABTS center dot+, and alpha-glucosidase inhibitory activity of buckwheat were considerably enhanced after processing by SSF with E. cristatum. Additionally, solvents with different polarities significantly influenced the antioxidant and alpha-glucosidase inhibitory activities of buckwheat extracts. Our study indicated that processing by SSF with E. cristatum can greatly improve the phytochemical components of buckwheat and consequently contribute to its antioxidant and alpha-glucosidase inhibitory activities. SSF with E. cristatum is an innovative method for enhancing the health-promoting components and bioactivities of buckwheat.

Current pharmacological therapies for the management of chronic articular diseases are far from being satisfactory, so new strategies need to be investigated. We tested the intra-articular pain relieving properties of a system of molecules from a characterizedCentella asiaticaextract (14G1862) in a rat model of osteoarthritis induced by monoiodoacetate (MIA). 14G1862 (0.2-2 mg mL(-1)) was intra-articularly (i.a.) injected 7 days after MIA, behavioural and histological evaluations were performed 14, 30 and 60 days after treatments. Moreover, the effect of 14G1862 on nitrate production and iNOS expression in RAW 264.7 macrophages stimulated with LPS was assessed. In vitro, 14G1862 treatment attenuated LPS-induced NO production and iNOS expression in a comparable manner to celecoxib. In vivo, 14G1862 significantly reduced mechanical allodynia and hyperalgesia, spontaneous pain and motor alterations starting on day 14 up to day 60. The efficacy was higher or comparable to that evoked by triamcinolone acetonide (100 mu g i.a.) used as reference drug. Histological evaluation highlighted the improvement of several morphological parameters in MIA + 14G1862-treated animals with particularly benefic effects on joint space and fibrin deposition. In conclusion, i.a. treatment withCentella asiaticais a candidate to be a novel effective approach for osteoarthritis therapy.

Hypochlorous acid (HOCl) is typically assumed to be the primary reactive species in free available chlorine (FAC) solutions. Lately, it has been shown that less abundant chlorine species such as chlorine monoxide (Cl2O) and chlorine (Cl-2) can also influence the kinetics of the abatement of certain organic compounds during chlorination. In this study, the chlorination as well as bromination kinetics and mechanisms of 12 olefins (including 3 aliphatic and 9 aromatic olefins) with different structures were explored. HOCl shows a low reactivity towards the selected olefins with species-specific second-order rate constants < 1.0 M(-1)s(-1), about 4-6 orders of magnitude lower than those of Cl2O and Cl-2. HOCl is the dominant chlorine species during chlorination of olefins under typical drinking water conditions, while Cl2O and Cl-2 are likely to play important roles at high FAC concentration near circum-neutral pH (for Cl2O) or at high Cl- concentration under acidic conditions (for Cl-2). Bromination of the 12 olefins suggests that HOBr and Br2O are the major reactive species at pH 7.5 with species-specific second-order rate constants of Br2O nearly 3-4 orders of magnitude higher than of HOBr (ranging from 0.01 to 10 3 M(-1)s(-1)). The reactivities of chlorine and bromine species towards olefins follow the order of HOCl < HOBr < Br2O < Cl2O approximate to Cl-2. Generally, electron-donating groups (e.g., CH2OH and CH3-) enhances the reactivities of olefins towards chlorine and bromine species by a factor of 3-10(2) , while electron- withdrawing groups (e.g., Cl-, Br-, NO2-, COOH-, CHO-,-COOR, and CN-) reduce the reactivities by a factor of 3-10(4). A reasonable linear free energy relationship (LFER) between the species-specific second-order rate constants of Br2O or Cl2O reactions with aromatic olefins and their Hammett sigma(+) was established with a more negative rho value for Br2O than for Cl2O, indicating that Br2O is more sensitive to substitution effects. Chlorinated products including HOCl-adducts and decarboxylated Cl-adduct were identified during chlorination of cinnamic acid by high-performance liquid chromatography/high resolution mass spectrometry (HPLC/HRMS). As one of the most popular beverages globally, tea has enormous economic, cultural, and medicinal importance that necessitates a comprehensive metabolomics study of this species. In this study, a large-scale targeted metabolomics analysis on two types of leaf tissues of nine tea cultivars from five representative geographical origins within China was carried out using the liquid chromatography-mass spectrometry technique. RNA-seq-based transcriptomic analysis was in parallel conducted on the same samples, and gene expression and metabolic differentiation between tissues as well as between the multiple tea cultivars were investigated. The data obtained provide an accessible resource for further studies of naturally occurring metabolic variation of tea plants, which will aid in thoroughly interpreting the underlying genetic and molecular mechanisms of biosynthesis of specialized metabolites in this critical species. Candidate genes including a transcription factor (CsMYB5-like), which were highly correlated with both the content of flavonoids and the expression level of genes participating in the phenylpropanoid and flavonoid biosynthesis pathway, were identified as potential targets for quality improvement of tea.

The fruits of Zanthoxylum armatum DC. were collected from different altitudes (1000-2000 m) and populations (wild and cultivated) from Salyan district, Nepal. The essential oil was extracted by Clevenger apparatus and the components were analyzed through GC-MS. The yield of essential oil obtained from hydro-distillation of fruits ranged from 2.72 to 7.6%. The maximum yield was 7.6% from wild fruits at 1600-1800 m altitude and the minimum was 2.72% from cultivated fruits at 1000-1200 m altitude. This was the highest recorded essential oil yield from Z. armatum fruits. A total of 13 volatile compounds were identified from the essential oil by GC-MS analysis. The major components were linalool, cinnamate (E)methyl, limonene, myrcene, sabinene and terpinen-4-ol, which were present in higher proportion in all the samples. Other components were identified in a very low proportion. The main component linalool occurred in the highest proportion (74.12%) from wild populations at 1600-1800 m altitude and the lowest (44.73%) was from cultivated populations at 1000-1200 m altitude. The highest proportion of linalool was also reported for the first time in this study from Z. armatum fruits. Among the six major components of the essential oil, terpinen-4-ol was present in the lowest proportion. Results of the present study indicated that the altitude and habitat types could affect the essential oil composition in Zanthoxylum armatum fruits.

Electronic cigarettes (ECs) are categorized into generations which differ in terms of design, aerosol production, and customizability. Current and former smokers prefer third-generation devices that satisfy tobacco cravings more effectively than older generations. Recent studies indicate that EC aerosols from first- and second-generation devices contain reactive carbonyls and free radicals and can cause in vitro cytotoxicity. Third-generation ECs have not been adequately studied. Further, previous studies have focused on cells from the respiratory tract, whereas those of the oral cavity, which is exposed to high levels of EC aerosols, have been understudied. We quantified the production of reactive carbonyls and free radicals by a third-generation EC and investigated the induction of cytotoxicity and oxidative stress in normal and cancerous human oral cell lines using a panel of eight commercial EC liquids. We found that EC aerosols produced using a new atomizer contained formaldehyde, acetaldehyde, and acrolein, but did not contain detectable levels of free radicals. We found that EC aerosols generated from only one of the eight liquids tested using a new atomizer induced cytotoxicity against two human oral cells in vitro. Treatment of oral cells with the cytotoxic EC aerosol caused a concomitant increase in intracellular oxidative stress. As atomizer age increased with repeated use of the same atomizer, carbonyl production, radical emissions, and cytotoxicity increased. Overall, our results suggest that thirdgeneration ECs may cause adverse effects in the oral cavity and normal EC use, which involves repeated use of the same atomizer to generate aerosol, may enhance the potential toxic effects of third-generation ECs.
