A methylation reagent for compounds bearing N-H, O-H, and S-H functionalities was developed. Methyl trifluoroacetate (MTFA) was commonly considered as trifluoroacetylating reagent or trifluoromethylating reagent. In this work, we report the methylation behavior of MTFA under mild conditions with good functional group tolerance, allowing the transformation of a wide range of substrates, including N,H-heteroaromatic compounds, phenolic compounds, carboxylic acids, thiophenols, secondary amides and imides, in high yields. This method was preliminarily applied to the chemoselective methylation of bifunctionalized secondary amide.

Using thianthrene S-oxide (TTSO) as a transient mediator, para-arylation and alkenylation of mono-substituted arenes have been demonstrated via a para-selective thianthrenation/Pd-catalyzed thio-Suzuki-Miyaura coupling sequence under mild conditions. This reaction features a broad substrate scope, and functional group and heterocycle tolerance. The versatility of this approach was further demonstrated by late-stage functionalization of complex bioactive scaffolds, and direct synthesis of some pharmaceuticals, including Tetriprofen, Ibuprofen, Bifonazole, and LJ570.

We report an inversed Arbuzov reaction of the fullerene derivatives C60Ar5Cl with trialkyl phosphites P(OR)(3) producing alkylated fullerene derivatives C60Ar5R (R = Me, Et, iPr, nBu) with almost quantitative yields. This reaction provides a convenient synthetic route for the preparation of a large variety of functionalized fullerene derivatives with tailored properties, e.g. water-soluble compounds demonstrating promising antiviral activities against HCMV, HSV1, HIV and several influenza virus strains.

Background: Nanotechnology-based strategies in the treatment of cancer have potential advantages because of the favorable delivery of nanoparticles into tumors through porous vasculature. Materials and Methods: In the current study, we synthesized a series of water-soluble fullerene derivatives and observed their anti-tumor effects on human lung carcinoma A549 cell lines. The quantitative structure-activity relationship (QSAR) modeling was employed to investigate the relationship between anticancer effects and descriptors relevant to peculiarities of molecular structures of fullerene derivatives. Results: In the QSAR regression model, the evaluation results revealed that the determination coefficient r(2) and leave-one-out cross-validation q(2) for the recommended QSAR model were 0.9966 and 0.9246, respectively, indicating the reliability of the results. The molecular modeling showed that the lack of chlorine atom and a lower number of aliphatic single bonds in saturated hydrocarbon chains may be positively correlated with the lung cancer cytotoxicity of fullerene derivatives. Synthesized water-soluble fullerene derivatives have potential functional groups to inhibit the proliferation of lung cancer cells. Conclusion: The guidelines obtained from the QSAR model might strongly facilitate the rational design of potential fullerene-based drug candidates for lung cancer therapy in the future.

The N-heterocyclic carbene-catalyzed beta-addition of enals to 3-alkylenyloxindoles was developed. We report high-performance I+/H(2)O(2)catalysis for the oxidative or decarboxylative oxidative alpha-azidation of carbonyl compounds by using sodium azide under biphasic neutral phase-transfer conditions. To induce higher reactivity especially for the alpha-azidation of 1,3-dicarbonyl compounds, we designed a structurally compact isoindoline-derived quaternary ammonium iodide catalyst bearing electron-withdrawing groups. The nonproductive decomposition pathways of I+/H(2)O(2)catalysis could be suppressed by the use of a catalytic amount of a radical-trapping agent. This oxidative coupling tolerates a variety of functional groups and could be readily applied to the late-stage alpha-azidation of structurally diverse complex molecules. Moreover, we achieved the enantioselective alpha-azidation of 1,3-dicarbonyl compounds as the first successful example of enantioselective intermolecular oxidative coupling with a chiral hypoiodite catalyst.
