Dan-Huang-Qu-Yu capsule, a Chinese herbal medicine compound preparation, is widely used for chronic pelvic inflammatory disease. In this study, a rapid, selective, and sensitive microwave-assisted extraction ultra-high-performance liquid chromatography-Q Exactive quadrupole-orbitrap high-resolution mass spectrometry method was developed for analyzing its chemical compositions. A total of 85 compounds, including 22 flavonoids, 8 terpenoids, 5 quinones, 5 phthaleolactone, 23 organic acids, and 22 other compounds were identified from Dan-Huang-Qu-Yu capsule. Among them, 35 major compounds were unambiguously detected by comparing them with reference standards and selected as quality control markers, which were simultaneously determined in Dan-Huang-Qu-Yu capsule. The established method was successfully validated and applied for simultaneous determination of 35 bioactive compounds in Dan-Huang-Qu-Yu capsule from ten sample batches. The quantitative data of the analytes were analyzed by principal component analysis for quality assessment of Dan-Huang-Qu-Yu capsule. Six compounds (e. g., astragaloside IV, salvianolic acid B, ellagic acid, chlorogenic acid, N-butylidenephthalide, and luteolin) were screened out and regarded as chemical markers for quality control of Dan-Huang-Qu-Yu capsule. The established method has been proved to be a novel and useful tool for rapid research of Dan-Huang-Qu-Yu capsule. This research will provide reference for the scientific research of traditional Chinese medicines.

A new triazole-functionalized tetracarboxylic acid ligand (H4L) has been synthesized and utilized for the fabrication of a 3D Zn-II organic framework with a Zn-4(-COO)(6) cluster as the secondary building unit. The framework exhibits very good thermal stability and consists of dual functionalities of exposed Lewis acidic metal sites and accessible nitrogen-donor Lewis basic sites. Conventional methods to improve the flame retardancy of polymeric materials usually involve the use of flame-retardant elements such as Cl, Br and P, however, their use may bring more smoke and toxic gases hazards, and more importantly, cause non-negligible environmental and ecological problems. In this work, we put forward a novel green strategy that eliminates the use of any conventional flame-retardant elements to improve the flame retardancy by incorporating a synergistically cross-linkable structure (named PN) containing phenylacetylene and phenylimide groups. The resulting PN copolymer exhibited an excellent 55% lower smoke release rate and 68% lower heat release rate than the pure polymer, as well as a high LOI value of 32% and UL-94 V-0 rating with excellent anti-dripping performance. TG-DSC, rheological and FTIR results proved the high cross-linking ability of the PN copolymer due to the synergistic cross-linking effect between the imide-isoimide rearrangement of phenylimide and the self-cross-linking of phenylacetylene. The SEM, Raman and Py-GC/MS results further upheld the condensed phase flame-retardant mechanism. This eco-friendly synergistic cross-linking strategy provided new perspective for the design and synthesis of polymeric materials with excellent flame retardancy, great anti-dripping performance, and low release of heat, smoke, and toxic gas.

Ovarian cancer is associated with a high percentage of recurrence of tumors and resistance to chemotherapy. Cancer stem cells (CSCs) are responsible for cancer progression, tumor recurrence, metastasis, and chemoresistance. Thus, developing CSC-targeting therapy is an urgent need in cancer research and clinical application. In an attempt to achieve potent and selective anti-CSC agents, a series of celastrol derivatives with cinnamamide chains were synthesized and evaluated for their anti-ovarian cancer activities. Most of the compounds exhibited stronger antiproliferative activity than celastrol, and celastrol derivative 7g with a 3,4,5-trimethoxycinnamamide side chain was found to be the most potent antiproliferative agent against ovarian cancer cells with an IC50 value of 0.6 mu M. Additionally, compound 7g significantly inhibited the colony formation ability and reduced the number of tumor spheres. Furthermore, compound 7g decreased the percentage of CD44(+), CD133(+) and ALDH(+) cells. Thus, compound 7g is a promising anti-CSC agent and could serve as a candidate for the development of new antiovarian cancer drugs. (C) 2019 Elsevier Masson SAS. All rights reserved.

Tea is an important beverage source of dietary polyphenols and well known for containing phenolic structure diversity. A series of phenylpropanoid-substituted catechins, flavonols, flavan-3-hexoside, and proanthocyanidin are present in different herbs with various biological activities, inspiring our exploration of phenylpropanoid-substituted ester type of catechins (PSECs) due to the enrichment of galloylated catechins in tea. In this study, we used a guiding-screening-location-isolation integrated route including creating a hypothesized PSEC dataset, MS/MS data acquiring, construction of molecular networks, and traditional column chromatography and preliminarily identified 14 PSECs by MS/MS spectrum. Two of these PSECs were further purified and elucidated by NMR and CD spectra. Further MS detection in tea products and fresh leaves suggests that the production of the two new compounds was enhanced during tea processing. The synthesis mechanism was proposed to obtain these types of components for further investigation on their roles in human health protection. This study provides an example for the exploration of new functional ingredients from food sources guided by MS/MS data-based networking, and also new insights into the reaction mechanism to form new catechin conjugates among polyphenols in green tea.

Current pharmacological therapies for the management of chronic articular diseases are far from being satisfactory, so new strategies need to be investigated. We tested the intra-articular pain relieving properties of a system of molecules from a characterizedCentella asiaticaextract (14G1862) in a rat model of osteoarthritis induced by monoiodoacetate (MIA). 14G1862 (0.2-2 mg mL(-1)) was intra-articularly (i.a.) injected 7 days after MIA, behavioural and histological evaluations were performed 14, 30 and 60 days after treatments. Moreover, the effect of 14G1862 on nitrate production and iNOS expression in RAW 264.7 macrophages stimulated with LPS was assessed. In vitro, 14G1862 treatment attenuated LPS-induced NO production and iNOS expression in a comparable manner to celecoxib. In vivo, 14G1862 significantly reduced mechanical allodynia and hyperalgesia, spontaneous pain and motor alterations starting on day 14 up to day 60. The efficacy was higher or comparable to that evoked by triamcinolone acetonide (100 mu g i.a.) used as reference drug. Histological evaluation highlighted the improvement of several morphological parameters in MIA + 14G1862-treated animals with particularly benefic effects on joint space and fibrin deposition. In conclusion, i.a. treatment withCentella asiaticais a candidate to be a novel effective approach for osteoarthritis therapy.

The practical problems of insufficient mechanical strength, poor processability, tedious synthesis and flammability facing shape memory polymers have limited their further usage in industrial fields, especially in construction and aerospace. A novel and very simple method of ternary polymerization is presented here to prepare multi-functional copolyesters from general poly(ethylene terephthalate) (PET) through the new well-designed third monomer that features pendent phenylacetylene-phenylimide units. The pi-pi stacking between phenylacetylene groups as a reversible net-point not only endows the copolyesters with good shape memory and self-healability, but also reinforces the interchain interaction, leading to high tensile strength (79.6-89.6 MPa). Interestingly enough, the phenylacetylene can synergistically crosslink with the unsaturated C N group generated from the phenylimide during burning, resulting in excellent flame retardancy. Thermoplastic smart copolyesters can be designed into fire alarms and can also be used for 3D printing. The printed geometries exhibit good shape memory behaviors, which could be deformed into a small size to save space during transportation/storage and allow manufacturing freedom for space applications.
