Hepatitis C virus (HCV) is a major global public health problem. While the current standard of care, a direct-acting antiviral (DAA) protease inhibitor taken in combination with pegylated interferon and ribavirin, represents a major advancement in recent years, an unmet medical need still exists for treatment modalities that improve upon both efficacy and tolerability. Toward those ends, much effort has continued to focus on the discovery of new DAAs, with the ultimate goal to provide interferon-free combinations. The RNA-dependent RNA polymerase enzyme NS5B represents one such DAA therapeutic target for inhibition that has attracted much interest over the past decade. Herein, we report the discovery and optimization of a novel series of inhibitors of HCV NS5B, through the use of structure-based design applied to a fragment-derived starting point. A review. Di-Me fumarate (DMF) is a first line medication for multiple sclerosis. It has a favorable safety profile, however, there is concern regarding the occurrence of moderate-severe and sustained lymphopenia and the associated risk of progressive multifocal leukoencephalopathy. We carried out an extensive literature review to understand the mol. mechanisms underlying this adverse reaction. Dynamic changes in certain components of the immune system are likely to be important for the therapeutic effects of DMF, including depletion of memory T cells and decrease in activated T cells together with expansion of naive T cells. Similar modifications were reported for the B cell components. CD8+ T cells are particularly susceptible to DMF-induced cell death, with marked reductions observed in lymphopenic subjects. The reasons underlying such increased sensitivity are not known, nor it is known how expansion of other lymphocyte subsets occurs. Understanding the mol. mechanisms underlying DMF action is challenging: in vivo DMF is rapidly metabolized to monomethyl fumarate (MMF), a less potent immunomodulator in vitro. Pharmacokinetics indicate that MMF is the main active species in vivo. However, the relative importance of DMF and MMF in toxicity remains unclear, with evidence presented in favor of either of the compounds as toxic species. Pharmacogenetic studies to identify genetic predictors of DMF-induced lymphopenia are limited, with inconclusive results.

A feeding trial was conducted to evaluate a new dietary protein Methanotroph (Methylococcus capsulatus, Bath) bacteria meal (BPM) on nutritional profile of Litopenaeus vannamei. The basal diet was formulated to contain 25% fish meal and then 15%, 30% and 45% of fish meal protein were replaced with BPM in three exptl. diets, which were labeled FM, BPM15, BPM30, and BPM45, resp. A total of 480 uniform-sized shrimp were equally distributed to four groups with three replicates and fed exptl. diets four times daily for seven weeks. Results showed that dietary BPM had no significant effect on the growth performance of shrimp (P > 0.05) but significantly reduced the crude lipid content of the whole body in shrimp fed the BPM30 diet compared to the FM diet (P < 0.05). Total cholesterol and triglyceride in the hemolymph fell significantly with the increase BPM substitution (P < 0.05). Expression of fas (fatty acid synthetase) significantly decreased with increasing BPM substitution, but the expression of cpt-1 (carnitine palmitoyltransferase) significantly increased in shrimp fed BPM30 (P < 0.05). Expression of tor (target of rapamycin) significantly decreased in hepatopancreas, while an opposite change was exhibited in the intestine of shrimp fed BPM45 (P < 0.05). Results of metabolomics indicated that dietary BPM affected the TCA cycle, ascorbate and aldarate metabolism, and glutathione metabolism in shrimp. In conclusion, BPM is an alternative to FM as a novel protein source for shrimp feed, but the changes in whole body composition, lipolysis and fatty acid synthesis, and protein synthesis and muscle metabolites of L.vannamei are noted. Dynamic urethane chem., which involves both associative and dissociative mechanisms, can be used to achieve reprocessability in cross-linked polyurethane (PU) networks. We discovered that property recovery and thermal stability of dynamic covalent PU networks can be enhanced by incorporating excess free hydroxyl groups, and thus performing polymerization reactions at slight stoichiometric imbalance of hydroxyl and isocyanate groups, and by increasing the crosslinker functionality from three to four. While free hydroxyl groups suppress the reversion of urethane links and minimize side reactions associated with liberated isocyanate groups under reprocessing conditions, tetrafunctional crosslinkers help to maintain network integrity in the presence of small levels of side reactions. Using these strategies, we developed a PU network that, within exptl. error, exhibits 100% recovery of crosslink d. and tensile properties after multiple reprocessing steps, which has not been reported before. We also demonstrated the potential of recovering monomer from PU networks by alcoholysis under relatively mild conditions. Our study not only offers a solution to the long-standing issue of PU network recycling, but also provides simple strategies to improve property recovery of reprocessable networks.

Sickle cell disease (SCD) is characterized by increased hemolysis, which results in plasma heme overload and ultimately cardiovascular complications. Here, we hypothesized that increased heme in SCD causes upregulation of heme oxygenase 1 (Hmox1), which consequently drives cardiomyopathy through ferroptosis, an iron-dependent non-apoptotic form of cell death. First, we demonstrated that the Townes SCD mice had higher levels of hemopexin-free heme in the serum and increased cardiomyopathy, which was corrected by hemopexin supplementation. Cardiomyopathy in SCD mice was associated with upregulation of cardiac Hmox1, and inhibition or induction of Hmox1 improved or worsened cardiac damage, resp. Because free iron, a product of heme degradation through Hmox1, has been implicated in toxicities including ferroptosis, we evaluated the downstream effects of elevated heme in SCD. Consistent with Hmox1 upregulation and iron overload, levels of lipid peroxidation and ferroptotic markers increased in SCD mice, which were corrected by hemopexin administration. Moreover, ferroptosis inhibitors decreased cardiomyopathy, whereas a ferroptosis inducer erastin exacerbated cardiac damage in SCD and induced cardiac ferroptosis in nonsickling mice. Finally, inhibition or induction of Hmox1 decreased or increased cardiac ferroptosis in SCD mice, resp. Together, our results identify ferroptosis as a key mechanism of cardiomyopathy in SCD.

Adequate nitrate availability throughout the strawberry life cycle improves fruit quality and productivity. However, the gene expression and metabolism patterns during nitrate-triggered developmental processes in fruits remain unknown. This transcriptome study reports the involvement of nitrate transporters and their correlated transcription factors (TFs) in the coordinate regulation of strawberry fruit development after nitrate treatment. Nitrate supply promoted plant growth and slightly delayed fruit ripening compared with the control. Nitrate also increased the contents of nitrogen, nitrogen-related metabolites, glucose, fructose, and sorbitol, but decreased the contents of several acid-related metabolites. Moreover, nitrate activated carbon and nitrogen metabolism at 25, 30, and 35 days post-anthesis (DPA). Nitrate treatment induced 10,206 differentially expressed genes (DEGs), which enriched carbon and nitrogen metabolism, amino acid, phenylpropanoid, flavonoid, terpenoid, carotenoid biosynthesis, and other metabolic pathways at 35 DPA. The DEGs also enriched GO terms for nitrate, nitrogen compound, and peptide transporter activities. Nitrate treatment significantly regulated 79 out of 164 nitrate transporter genes (FaNRTs) at 25, 30, and 35 DPA. Correlation anal. showed that strawberry adaptation to nitrate availability during fruit development involves dynamic multi-combinations between FaNRT and different TF families. qRT-PCR anal. showed that FaNPF6.3, FaNPF4.6, and FaNPF7.2 are potential nitrate signaling mols. and transporters for root nitrate uptake or removing nitrate from xylem vessels. Nitrate treatment induced FaNPF6.3, FaNPF4.6, and FaNPF7.2 expression while their correlated TFs (FaSPL8, FaNAC77-like, FaMYB1R1, and FaMYB44-like) transcription levels were down-regulated at 25 DPA. These results indicate that the spatio-temporal expression of nitrate transporters correlated with the increased nitrogen use efficiency (NUE) to enhance metabolism The results provide a fertilizer and water management strategy for improving quality and productivity in the strawberry industry.

The dendritic guest-host complexes showed a significantly higher emission upon binding then that of the individual mols. due to the three-dimensional orientation of the OPV guest mols. In the solid state, this enhancement in luminescence was a factor of 10. The π-conjugated oligomers are less aggregated in the supramol. assemblies presumably because of a shielding effect of the bulky adamantyl units present in the hosts.
