Short syntheses of (Z)-7-dodecen-1-yl acetate, (Z)-7-tetradec-1-yl acetate, (Z)-9-dodecenyl-1-yl acetate, and (Z)-9-tetradec-1-yl acetate use 7-hydroxyheptanal and 9-oxononanoic acid; precursors obtained by oxidative cleavage of aleuritic acid. The key step in these syntheses is a stereoselective Wittig reaction between aldehyde and alkylphosphonium salt. Wittig-Horner type reaction of 7-hydroxyheptanal and NCCH2P(O)(OEt)2 gave the α,β-unsaturated nitrile derivative which after protection of the OH group was reduced to the aldehyde. Wittig reaction of the latter, followed by acetylation, completed the synthesis of (E,Z)-7,9-dodecadien-1-yl acetate, the sex pheromone of the European grapevine moth, Lobesia botrana.

The recent detection of potent carcinogenic nitrosamine impurities in several human medicines has triggered product recalls and interrupted the supply of critical medications for hundreds of millions of patients, illuminating the need for increased testing of nitrosamines in pharmaceutical products. However, the development of anal. methods for nitrosamine detection is challenging due to high sensitivity requirements, complex matrixes, and the large number and variety of samples requiring testing. Herein, we report an anal. method for the anal. of a common nitrosamine, N-nitrosodimethylamine (NDMA), in pharmaceutical products using full evaporation static headspace gas chromatog. with nitrogen phosphorous detection (FE-SHSGC-NPD). This method is sensitive, specific, accurate, and precise and has the potential to serve as a universal method for testing all semi-volatile nitrosamines across different drug products. Through elimination of the detrimental headspace-liquid partition, a quantitation limit of 0.25 ppb is achieved for NDMA, a significant improvement upon traditional LC-MS methods. The extraction of nitrosamines directly from solid sample not only simplifies the sample preparation procedure but also enables the method to be used for different products as is or with minor modifications, as demonstrated by the anal. of NDMA in 10+ pharmaceutical products. The in situ nitrosation that is commonly observed in GC methods for nitrosamine anal. was completely inhibited by the addition of a small volume solvent containing pyrogallol, phosphoric acid, and isopropanol. Employing simple procedures and low-cost instrumentation, this method can be implemented in any anal. laboratory for routine nitrosamine anal., ensuring patient safety and uninterrupted supply of critical medications.

Three-dimensional (3D) cultures, so-called organoids, have emerged as an attractive tool for disease modeling and therapeutic innovations. Here, we aim to determine if boundary cap neural crest stem cells (BC) can survive and differentiate in gelatin-based 3D bioprinted bioink scaffolds in order to establish an enabling technol. for the fabrication of spinal cord organoids on a chip. BC previously demonstrated the ability to support survival and differentiation of co-implanted or co-cultured cells and supported motor neuron survival in excitotoxically challenged spinal cord slice cultures. We tested different combinations of bioink and crosslinked material, analyzed the survival of BC on the surface and inside the scaffolds, and then tested if human iPSC-derived neural cells (motor neuron precursors and astrocytes) can be printed with the same protocol, which was developed for BC. We showed that this protocol is applicable for human cells. Neural differentiation was more prominent in the peripheral compared to central parts of the printed construct, presumably because of easier access to differentiation-promoting factors in the medium. These findings show that the gelatin-based and enzymically crosslinked hydrogel is a suitable bioink for building a multicellular, bioprinted spinal cord organoid, but that further measures are still required to achieve uniform neural differentiation.

Following up the open initiative of anti-malarial drug discovery, a GlaxoSmithKline (GSK) phenotypic screening hit was developed to generate hydroxyethylamine based plasmepsin (Plm) inhibitors exhibiting growth inhibition of the malaria parasite Plasmodium falciparum at nanomolar concentrations Lead optimization studies were performed with the aim of improving Plm inhibition selectivity vs. the related human aspartic protease cathepsin D (Cat D). Optimization studies were performed using Plm IV as a readily accessible model protein, the inhibition of which correlates with anti-malarial activity. Guided by sequence alignment of Plms and Cat D, selectivity-inducing structural motifs were modified in the S3 and S4 sub-pocket occupying substituents of the hydroxyethylamine inhibitors. This resulted in potent anti-malarials with an up to 50-fold Plm IV/Cat D selectivity factor. More detailed investigation of the mechanism of action of the selected compounds revealed that they inhibit maturation of the P. falciparum subtilisin-like protease SUB1, and also inhibit parasite egress from erythrocytes. Our results indicate that the anti-malarial activity of the compounds is linked to inhibition of the SUB1 maturase plasmepsin subtype Plm X.

Finally, camouflage coating applications for encryption display films were successfully demonstrated.

The filling factor of LCs in the LC-polymer composite system is a crucial factor for the electro-optic performance of optically isotropic liquid crystal (OILC) devices. The polymerization-induced phase separation method is well-known method to fabricate the OILCs; however, this method spontaneously limits the filling factor by ∼ 40%, which is inadequately low, owing to low-LC concentration in the mixtures To overcome such intrinsic problem, we propose an encapsulated LC which is fabricated by means of oil-in-water encapsulation. Through this approach, the filling factor of LCs is effectively increased up to 67.8% because the capsules are closely packed. Consequently, we achieve a thin-composite film of nano-size LC encapsulation whose isotropic state is reliably switched to anisotropic state in response to an external elec. field with low-operation voltage (Eth = 1.4 V/μm and Eop = 4.6 V/μm), fast-response time (τon = 0.5 ms, τoff = 2.2 ms), and hysteresis-free electro-optic behavior. The highly improved electro-optic performances of the proposed encapsulated LC device would give versatile applications in blooming free-form flexible displays and various tunable photonic systems.

Reactivity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical in methanol, ethanol, isopropanol, isooctane, and Et acetate, was evaluated to assess the antioxidant capabilities in medium chain triacylglycerol. DPPH loss values were obtained over 30 min, with sampling every 5 min. Even the same concentration of antioxidants showed different DPPH reactivity depending on solvent. In methanol, 5 min was enough for α-tocopherol to react with DPPH, whereas BHT did not react with DPPH even after 30 min. Gallate series showed higher DPPH reactivity than TBHQ, sesamol, or BHA in methanol, while lower reactivity in isooctane. Antioxidants in ethanol and isopropanol reacted with DPPH less efficiently compared to those in methanol, the exception being sesamol. DPPH reactivity of gallate series in isooctane was lower than that of sesamol, TBHQ, and α-tocopherol. Combinatorial usage of methanol and isooctane for DPPH reactivity could provide reliable information on the antioxidant capacities of chems. in edible oils.

With rapid technol. advancements, efficient thermal management is becoming increasingly important to sustain the stable operation of electronic devices. In this study, aluminum nitride (AlN) fillers with various acrylate monomers were subjected to two types of silane surface treatments to prepare composites with a high loading of AlN filler (65 wt%). The acrylates-isobornyl acrylate (IBOA), 1,4-butanediol diacrylate (BDDA), and trimethylolpropane triacrylate (TMPTA)-were mixed with bisphenol A ethoxylate dimethacrylate (Bis-EMA) as an oligomer, and phenylbis (2,4,6-trimethylbenzoyl)phosphine oxide (BAPO) as a photo-initiator in different proportions to obtain resin matrixes. Pristine AlN and AlN functionalized with APTES and MPS were used as fillers. The effect of the acrylate functional group in silanes on the thermal and mech. properties of the acrylate resin was explored. The thermal conductivities of the IBOA/AlN/APTES and IBOA/AlN/TMPTA composites with a high loading of the filler functionalized with APTES and MPS were 1.34 and 1.57 W/(m•K), resp., 4.15 and 5.28 times higher than that of the composite with neat resin. The enhanced filler-matrix compatibility increased the tensile strength of the composites. The findings highlighted that silane functionalization of AlN can enhance the thermal conductivity and mech. properties of the composite.

Korean traditional rice wines yakju and takju are generally brewed with nuruk as the source of the saccharogenic enzymes by natural fermentation To improve the quality of Korean rice wine, the microorganisms in the nuruk need to be studied. The objective of this research was to improve the quality of Korean wine with the wild-type yeast strains isolated from the fermentation starter, nuruk. Only strain YA-6 showed high activity in 20% ethanol. Precipitation of Y89-5-3 was similar to that of very flocculent yeast (>80%) at 75.95%. Using 18S rRNA sequencing, all 10 strains were identified as Saccharomyces cerevisiae. Volatile compounds present in yakju were analyzed by gas chromatog.-mass selective detector. The principal component anal. (PCA) of the volatile compounds grouped long-chain esters on the right side of the first principal component, PC1; these compounds were found in yakju that was made with strains YA-6, Y89-5-3, Y89-5-2, Y90-9, and Y89-1-1. On the other side of PC1 were short-chain esters; these compounds were found in wines that were brewed with strains Y183-2, Y268-3, Y54-3, Y98-4, and Y88-4. Overall, the results indicated that using different wild-type yeast strains in the fermentation process significantly affects the chem. characteristics of the glutinous rice wine.
