Cholesterol decreases the activity of the majority of ion channels while increasing the activity of only a few, yet it remains unclear how. Here, we used the inwardly rectifying potassium channel Kir3.4, which is up-regulated by cholesterol, as a tool to address this question. Employing mutagenesis and electrophysiol., we discovered a mol. switch that controls the impact of cholesterol on the channel. Through a single point mutation at position 182 in the transmembrane domain of Kir3.4, we converted the cholesterol-driven up-regulation of the channel into down-regulation. Microseconds-long coarse-grained and atomistic mol. dynamics simulations revealed that the effect of the point mutation propagated toward the selectivity filter of the channel whose conformation controls the conductance of the channel. Planar lipid bilayer experiments validated these results, showing that although cholesterol up-regulated Kir3.4 by increasing its open probability, cholesterol down-regulated the mutant by decreasing its conductance. In this study, an environmentally friendly and non-toxic route to synthesize lignin-based non-isocyanate poly(imine-hydroxyurethane)s networks was explored. Specifically, the NH2-terminated polyhydroxyurethanes (NPHUs) prepolymer was first synthesized from bis(6-membered cyclic carbonate) (BCC) and diamine via the ring-opening reaction. The structural, mech., and thermal properties of NIPUs with different stoichiometric feed ratios of BCC and levulinate lignin were characterized by Fourier transform IR spectroscopy, NMR, differential scanning calorimetry, dynamic mech. anal., and thermogravimetric anal.. The results indicated that the tensile strength, Young’s modulus, toughness, storage modulus, glass transition temperature, and thermal stability of lignin-based NIPUs gradually increased with increasing lignin content, and the highest Young’s modulus of 41.1 MPa was obtained when lignin content reached 45.53%. With good reprocessing properties, this synthetic framework of lignin-based NIPUs also provides sustainable non-isocyanate-based substitutions to traditional polyurethane networks. According to the findings of this study, G/SHMP/OEO films could be a promising tool for packaging and protecting meat-based foods. Nevertheless, current mRNA/LNP vaccine technol. has still some limitations, one of them being thermostability, as evidenced by the ultracold distribution chain required for the licensed vaccines. We found that the thermostability of mRNA/LNP, could be improved by a novel imidazole modified lipid, DOG-IM4, in combination with standard helper lipids. DOG-IM4 comprises an ionizable head group consisting of imidazole, a dioleoyl lipid tail and a short flexible polyoxyethylene spacer between the head and tail. Here we describe the synthesis of DOG-IM4 and show that DOG-IM4 LNPs confer strong immunization properties to influenza HA mRNA in mice and macaques and a remarkable stability to the encapsulated mRNA when stored liquid in phosphate buffered saline at 4°C. We speculate the increased stability to result from some specific attributes of the lipid’s imidazole head group. Lipid nanoparticles (LNPs) are the leading technol. for RNA delivery, given the success of the Pfizer/BioNTech and Moderna COVID-19 mRNA vaccines, and small interfering RNA (siRNA) therapies (patisiran). However, optimization of LNP process parameters and compositions for larger RNA payloads such as self-amplifying RNA (saRNA), which can have complex secondary structures, have not been carried out. Furthermore, the interactions between process parameters, critical quality attributes (CQAs), and function, such as protein expression and cellular activation, are not well understood. Here, we used two iterations of design of experiments (DoE) (definitive screening design and Box-Behnken design) to optimize saRNA formulations using the leading, FDA-approved ionizable lipids (MC3, ALC-0315, and SM-102). We observed that PEG is required to preserve the CQAs and that saRNA is more challenging to encapsulate and preserve than mRNA. We identified three formulations to minimize cellular activation, maximize cellular activation, or meet a CQA profile while maximizing protein expression. The significant parameters and design of the response surface modeling and multiple response optimization may be useful for designing formulations for a range of applications, such as vaccines or protein replacement therapies, for larger RNA cargoes. Morinda citrifolia has aroused the interest of several research institutions due to its pharmacol. properties. The compounds biosynthesized can be explored as an alternative for control measures of plant pathogens causing leaf lesions in maize. The study aim at evaluating the potential effects of essential oil obtained from ripe fruits of M. citrifolia on Bipolaris maydis and Exserohilum turcicum isolated from maize plants.
