The recent growing concern driven by consumer interest in the safety and quality of seafood, has boosted the search for healthy and functional aquafeeds. The current study represents the first approach to assess the potential of volatile composition of the wine industry by products (e.g., grape pomace, grape stems, lees), as additives for improving the quality of fish feeds in terms of organoleptic characteristics (e.g., aroma and flavor) and health benefits. Headspace solid-phase microextraction followed by gas chromatog.-mass spectrometry (HS-SPME/GC-MS) was used to establish the volatile profile of wine industry byproducts. A total of 153 volatile organic compounds (VOCs), which belong to different chem. families, comprising 36 esters, 31 carbonyl compound, 20 alcs., 18 terpenoids, 17 acids, 11 furanic compounds, four volatile phenols, two lactones, and 14 miscellaneous, were identified. Esters and terpenoids showed a pos. contribution to the aquafeeds aroma with fruity, sweet, green, fresh, and berry notes, whereas some acids (e.g., hexanoic acid) and terpenoids (e.g., limonene) could be used as antimicrobial, antioxidant and antiproliferative agents. Our findings confirmed the potential of wine industry byproducts as a rich source of essential compounds to enhance the quality of aquafeeds towards the valorization of winery waste based on the concept of circular economy. Further investigation on the extraction, isolation and purification of VOCs from a natural bio-source will guarantee the safety of the aquafeed and compliance with the requirements of the animal feed industry. Acetyl xylan esterases (AXEs) are enzymes capable of hydrolyzing the acetyl bonds in acetylated xylan, allowing for enhanced activity of backbone-depolymerizing enzymes. In this article, we report the characterization of a novel AXE identified as Gene_id_40363 in the metagenomic library analyzed from the gut microbiota of the common black slug. The conserved domain description was identified with an NCBI BLASTp search using the translated nucleotide sequence as a query. The activity of the recombinant enzyme was tested on various synthetic substrates and acetylated substrates. The protein sequence matched the conserved domain described as putative hydrolase and aligned closely to an uncharacterized esterase from Buttiauxella agrestis, hence the designation as BaAXE. BaAXE showed low sequence similarity among characterized CE family proteins with an available 3D structure. BaAXE was active on 4-nitrophenyl acetate, reporting a specific activity of 78.12 U/mg and a Km value of 0.43 mM. The enzyme showed optimal activity at 40°C and pH 8 and showed high thermal stability, retaining over 40% activity after 2 h of incubation from 40°C to 100°C. BaAXE hydrolyzed acetyl bonds, releasing acetic acid from acetylated xylan and β-D-glucose pentaacetate. BaAXE has great potential for biotechnol. applications harnessing its unique characteristics. Bioactive substances possess superior antioxidant, anti-hypertensive, and anti-tumor activities. However, their susceptibility to degrade at low pH often limit their application. Compare with the conventional oil-in-water (W/O) emulsions, the lack of natural hydrophobic stabilizers resulted in difficulties in fabricating water-in-oil (W/O) high internal phase emulsion (HIPE). Further, the instability of W/O HIPE also limit its industrial application in food industry. In the present study, a natural, conventional, eco-friendly, and structured W/O HIPE was developed using glyceryl monostearate (GMS) as emulsifier and beeswax and gellan gum as structurant. The system with the synergetic effect of emulsifier and structurant, accounted for 0.25 wt % of GMS and hydrogel, and 1.25 wt % of beeswax in total system endowed the W/O HIPE with biphasic rigid network and impressive storage stability. The microstructures of these newly developed W/O HIPEs could be controlled by the beeswax concentration (cb), GMS concentration (cg), and aqueous phase volume (Φ). Moreover, the GMS-based emulsion encapsulation improved the retention rate of betanin by 36.68% after 7 days of storage. Meanwhile, this emulsion gel could prevent the nucleophilic attack of hydrogen ions against betanin at room temperature under a low pH environment, which greatly improved the stability of betanin under highly acid conditions. Furthermore, the GMS stabilized W/O HIPE possessed pH-sensitive, which could achieve controlled-release of both hydrophilic and hydrophobic functional compounds in an in vitro simulated release assay. mRNA based gene therapies hold the potential to treat multiple diseases with significant advantages over DNA based therapies, including rapid protein expression and minimized risk of mutagenesis. However, successful delivery of mRNA remains challenging, and clin. translation of mRNA therapeutics has been limited. This study investigated the use of a lipid/polymer hybrid (LPH) nanocarrier for mRNA, designed to address key delivery challenges and shuttle mRNA to targeted tissues. LPH nanocarriers were synthesized using a scalable microfluidic process with a variety of material compositions and mRNA loading strategies. Results show that a combination of permanently ionized and transiently, pH-dependent ionizable cationic lipids had a synergistic effect upon on mRNA gene translation, when compared to each lipid independently. Upon i.v. administration, particles with adsorbed mRNA outperformed particles with encapsulated mRNA for protein expression in the lungs and the spleen despite significant LPH nanoparticle localization to the liver. In contrast, encapsulated particles had higher localized expression when injected i.m. with protein expression detectable out to 12 days post injection. I.m. administration of particles with OVA mRNA resulted in robust humoral immune response with encapsulated outperforming adsorbed particles in terms of antibody titers at 28 days. The jujube tree, Ziziphus jujube Mill, is one of the most important economic fruits in north China. Japanese wax scale, Ceroplastes japonicus Green, is a destructive pest in the jujube orchard. This paper studied the changes of the volatile of the jujube trees that were induced with Methyl-Jasmonic (MeJA), an exogenous signaling material, or were attacked by adults of the scale insect, C. japonicus. The headspace volatile trapping instrument was used to collect volatiles from jujube trees. The chem. components of the collected volatiles were analyzed using chromatog./mass spectroscopy (GC/MS). The results showed that the volatile substances were changed both in the components and in emission quantity when the jujube trees treated with MeJA or damaged by the scale insect. Twenty-two components were examined from the volatile samples from the MeJA-treated jujube tree, and their emission quantity was 37347 ng/h. Meanwhile, 24 components were determined in the volatiles from the scale insect-damaged jujube trees and their emission quantity reached 54292 ng/h. In comparison, 14 compounds were detected only in the volatiles of the health trees (as control) and the total emission quantity was 18611 ng/h. The difference reached the significant level either between the MeJA-treated section and the control section or between the scale insect-damaged section and the control section.
