Stable mol. conformation and intermol. forces are essential for peptide self-assembly. In this study, one novel dehydropeptide (DDP) monomer (Boc-(Z)Cα,b-ΔPhe-Gly-NHMe, DDP 1) was prepared; its conformation was confirmed to be more stable than the normal peptide 2 by NMR (NMR) and X-ray crystal diffraction experiments DDP 1 was self-assembled to one novel dehydropeptide nanomaterial (DDPN 1). Fourier transform IR (FTIR) spectroscopy results showed that hydrogen bonding was the main driving force of self-assembly. Electron microscope images displayed that the DDPN 1 fibers were longer and more stable than peptide 2 nanomaterials. Results of cell activity and enzyme hydrolysis proved that DDPN 1 had excellent biocompatibility and resistance to the enzymic hydrolysis of protease K. Therefore, the DDPN 1 was used to load the antitumor drug temozolomide (TMZ). Due to intermol. hydrogen bonds formed between TMZ and DDPN 1, TMZ-loaded DDPN 1 had a high percent entrapment efficiency (EE) of 83.72 ± 4.30% (n = 8) and a percent drug loading efficiency (LE) of 6.70 ± 0.34% (n = 8), and the half-life of TMZ-loaded DDPN 1 was 2.5-3 times longer than that of TMZ at pH 7. The in vitro cell viability results revealed that TMZ-loaded DDPN 1 exhibited higher antitumor activity (IC50 = 552.1 μM) against U118-MG than that of TMZ (IC50 = 1980.1 μM), possibly because that U118-MG cells uptook more TMZ from TMZ-loaded DDPN 1 than from free TMZ directly. This study is expected to inspire the design of biocompatible nanocarriers applied for anti-enzymic hydrolysis in drug delivery systems.

Alpha linolenic acid is an essential unsaturated fatty acid in organisms. However, there is a large gap between α-linolenic acid accumulation and its synthesis mechanism in insects. Fatty acid synthases (FASs) and desaturases (Desats) are vital enzymes required for the synthesis of unsaturated fatty acids. The pupae of Glyphodes pyloalis (Lepidoptera: Pyralidae), which is a destructive pest of mulberry trees, contain the highest level of α-linolenic acid compared to other life-history stages. To further explore the synthesis mechanism of α-linolenic acid in G. pyloalis pupae, author constructed a pupal transcriptome dataset and identified 106 genes related to fatty acid metabolism from it. Among these, two fatty acid synthases (GpylFAS) and five desaturases (GpylDesat) were identified. A qRT-PCR validation revealed that GpylFAS1 and GpylDesat1, 2, 3, 5 were expressed highest at pupal stages. Furthermore, the content of α-linolenic acid decreased significantly after silencing GpylFAS1 and GpylDesat5, resp. Besides, knocking down GpylFAS1 or GpylDesat5 resulted in more malformed pupae and adults, as well as lower emergence rates. Meanwhile, silencing GpylFAS1 or GpylDesat5 affected the expressions of the other GpylFASs and GpylDesats. The present results illustrate the pivotal function of FASs and Desats in α-linolenic acid biosynthesis and metamorphosis in insects. Authors research also broadens the sources of unsaturated fatty acids, especially for α-linolenic acid from insects, and provides novel insights for the management of mulberry insect pests from the perspective of utilization rather than control.

The dynamic behaviors of vitrimer are influenced by the interaction of crosslink d. and d. of exchangeable groups, which results in the difficulty of extracting the effect of mono-variation. In this study, the dynamic behaviors of vitrimer were tuned by changing the d. of exchangeable hydroxyls alone, while leaving the crosslink d. undisturbed. A multi-hydroxy vitrimer network without a loading catalyst was synthesized successfully. Three-armed functionalized polycarbonate, obtained by ring-opening polymerization of 1,3-dioxan-2-one and its derivatives, was treated as a precursor polymer. Afterward, the precursor polymer was converted into vitrimer in a single step by treatment with hexamethylene diisocyanate. When the system with constant crosslink d. was established, the accurate effect of the hydroxyl d. on the dynamic behaviors could be characterized by stress relaxation. The relationship of essential dynamic behavioral parameters, such as topol. freezing transition temperature (Tv), exchange activation energy (Ea), and Arrhenius prefactor (τ0), with the d. of exchangeable hydroxyls was quant. discussed. The results indicated that Tv and Ea were neg. correlated with the d. of exchangeable hydroxyls, while τ0 showed a pos. correlation. In addition, linear models were developed to describe the relationship between dynamic behaviors of vitrimer and d. of exchangeable hydroxyls. It is expected that the findings can be applied to precisely control Ea, τ0, Tv, and characteristic relaxation time (τ*) of vitrimer and can also be used as a model for the synthesis of various vitrimer materials.

Data encryption, protection, and anticounterfeiting are crucial in the modern information society. Though a number of technologies were developed, data security is still facing severe challenges. Herein, a fundamental new approach capable of multilevel data encryption, protection, and anticounterfeiting is developed based on the responsive zwitterionic polymers named as poly(pentafluorophenyl acrylate)-co-poly[(3-((3-acrylamidopropyl)dimethylammonio)propane-1-sulfonate)] (pPFPA-co-pADPS). Detailed studies reveal that zwitterionic polymers exhibit drastic elec. property changes to humidity variation. By reliance of this feature, highly secure touchless palm-based biometric authentication is realized; further, field-effect transistors (FETs) using zwitterionic polymers as dielecs. show nonvolatile memory behaviors with good reliability and rewrite-ability. Remarkably, not only appropriate voltages but also specific humidity range are required to safely decrypt the stored message. In addition, the memory gains the ability to destroy the stored data under counterfeiting/hacking condition, thus preventing the data from being stolen by repeated trials. Therefore, a high level of data security is realized. This work provides new insights into zwitterionic copolymer dielec. based OFETs, demonstrates the first example of double-stage data encryption and protection by OFET memory, and opens up new opportunities for data-securing by responsive materials.

Compared with I as standard, 1-[[[[5-(4-nitrobenzene)thio]-2-furanyl]-methylene]amino]-2,4-imidazolidinedione exhibited higher activity, and 1-[[[[5-(4-nitrobenzene)ethenyl]-2-furanyl]-methylene]amino]-2,4-imidazolidinedione showed comparable activity at 0·5, 3 and 6 h after administration to mice. There is a correlation between the electronic transmission ability of the furan nucleus in all 3 compounds and muscle relaxant activity.

Screening inhibitors of flavin monooxygenase 3 (FMO3) is very important for treating trimethylamine N-oxide (TMAO) derived thrombotic diseases. Herein, focusing on Xuefu Zhuyu decoction (XFZYD) as a Chinese traditional medicine with antithrombotic efficacy, a facile and label-free fluorescence strategy was developed for evaluating the influence of the bioactive ingredients in XFZYD on FMO3 activity through indicator displacement assay. To this end, the optimized supramol. host-guest (p-sulfonatocalix[4]arene-oxazine 1) reporter pair and FMO3 catalytic system were exploited to determine the influence of the bioactive compounds in XFZYD on the conversion from TMA to TMAO. From the nine compounds tested, naringin, paeoniflorin, β-ecdysterone, 18β-glycyrrhizic acid, amygdalin, albiflorin, and saikosaponin A downregulated FMO3 activity and reduced TMAO biosynthesis. Moreover, mol. docking was successfully applied to simulate the optimal conformation of a receptor-ligand complex between FMO3 and all tested compounds except for β-ecdysterone. Therefore, this approach provides a novel and promising strategy for screening FMO3 inhibitors from Chinese traditional medicine by supramol. sensing.

Lithium-sulfur (Li-S) batteries are considered a promising next generation alternative to lithium-ion batteries for energy storage systems due to its high energy d. However, several challenges, such as the polysulfide redox shuttle causing self-discharge of the battery, remain unresolved. In this paper, we explore the use of polymer etched ion-track membranes as separators in Li-S batteries to mitigate the redox shuttle effect. Compared to com. separators, their unique advantages lie in their very narrow pore size distribution, and the possibility to tailor and optimize the d., geometry, and diameter of the nanopores in an independent manner. Various polyethylene terephthalate membranes with diameters between 22 and 198 nm and different porosities were successfully integrated into Li-S coin cells. The reported coulombic efficiency of up to 97% with minor reduction in capacity opens a pathway to potentially address the polysulfide redox shuttle in Li-S batteries using tailored membranes.
