Trout were exposed to 3 dietary concentrations of each chem. to investigate the relationship between dietary exposure concentration and observed accumulation and depuration. Both EHMC and OCT were significantly biotransformed, resulting in mean in vivo whole-body biotransformation rate constants (kMET) of 0.54 ± 0.06 and 0.09 ± 0.01 d-1, resp. The kMET values generated for both chems. did not differ between dietary exposure concentrations, indicating that chem. concentrations in the fish were not high enough to saturate biotransformation enzymes. Both somatic and luminal biotransformation substantially reduce EHMC and OCT bioaccumulation potential in trout. Biomagnification factors (BMFs) and bioconcentration factors (BCFs) of EHMC averaged 0.0035 kg lipid kg lipid-1 and 396 L kg-1, resp., whereas those of OCT averaged 0.0084 kg lipid kg lipid-1 and 1267 L kg-1. These values are 1 to 2 orders of magnitude lower than the BMFs and BCFs generated for reference chems. of similar log KOW. Following a request from the European Commission, the EFSA Panel on Additives and Products or Substances used in Animal Feed ( FEEDAP ) was asked to deliver a scientific opinion on the safety and efficacy of expressed mandarin oil from the fruit peels of Citrus reticulata Blanco, when used as a sensory additive (flavouring) in feed and water for drinking for all animal species. The FEEDAP Panel concluded that the essential oil under assessment is safe up to the maximum proposed use levels in complete feed of 15 mg/kg for poultry, 33 mg/kg for pigs, 30 mg/kg for ruminants, 40 mg/kg for horse, and 15 mg/kg for salmon and rabbit. The presence of perillaldehyde was identified as a source of potential concern. However, in target species fed citrus byproducts as part of daily feed the use of the expressed mandarin oil in feed was not expected to increase the exposure to perillaldehyde to a relevant extent (< 4%). For companion animals and ornamental fish not normally exposed to citrus byproducts, no conclusion can be drawn. The FEEDAP Panel considered that the use in water for drinking is safe provided that the total daily intake of the additive does not exceed the daily amount that is considered safe when consumed via feed. No concerns for consumer safety were identified following the use of the additive up to the maximum proposed use level in feed. The essential oil under assessment should be considered as irritant to skin, eyes and the respiratory tract, and as a skin sensitizer. The use of the additive in animal feed under the proposed conditions of use was not expected to pose a risk for the environment. Expressed mandarin oil was recognized to flavor food. Since its function in feed would be essentially the same as that in food, no further demonstration of efficacy was considered necessary. We herein report the design, synthesis, and functional impact of an okadaic acid (OA) small analog, ITH12680, which restores the activity of phosphoprotein phosphatase 2A (PP2A), whose deficient activity has been implicated in nicotine-mediated tumor progression and chemoresistance in non-small cell lung cancer (NSCLC). For its design, we paid attention to the structure of the PP2A-OA complex, where the C16-C38 OA fragment confers PP2A affinity and selectivity, but it is not involved in the inhibitory effect. Confirming this hypothesis, PP2A activity was not inhibited by ITH12680. By contrast, the compound partially restored OA-exerted PP2A inhibition in vitro. Moreover, flow cytometry and immunoblotting experiments revealed that ITH12680 reversed nicotine-induced cisplatin resistance in NSCLC cells, as it prevented nicotine-induced reduction of Bax expression and inhibited nicotine-mediated activation of cell survival and proliferation kinases, Akt and ERK1/2. Our findings suggest that the rescue of nicotine-inhibited PP2A activity could diminish the resistance to cisplatin treatment observed in NSCLC patients who continue smoking.
