A wider polyethersulfone (PES) membrane pore size was employed in POCIS exposed in a static calibration experiment to investigate the uptake of 21 emerging contaminants ranging from hydrophilic (perfluoroalkyl compounds, xanthines, an artificial sweetener) to more hydrophobic compounds (pharmaceuticals, estrogens, UV filters). Compared to standard POCIS with 0.1-μm pore size PES membranes, the POCIS with 5-μm pore size PES membranes did not increase sampling rates for compounds of relatively low and mid-hydrophobicity. However, the uptake of more hydrophobic and anionic compounds, which either poorly diffuse through or are retained within the standard 0.1-μm PES membrane, showed a marked increase. This led to the first ever recorded sampling rates for triclosan (0.249 L day-1) and two UV filters (0.075-0.123 L day-1). Based on these results, more attention should be placed on the choice of the appropriate membrane for each POCIS application. The most suitable configuration depends on the studied compound physico-chem. characteristics-such as the polarity and the compound membrane-to-sorbent partitioning coefficient-but also on the site conditions (deployment time, fouling, flow variations, et.). Dust samples were collected from central NC homes (n = 188), solvent extracted, and split into two fractions, one for anal. using three different bioassays (nuclear receptor activation/inhibition and adipocyte development) and one for mass spectrometry (targeted measurement of 124 organic contaminants, including flame retardants, polychlorinated biphenyls, perfluoroalkyl substances, pesticides, phthalates, and polycyclic aromatic hydrocarbons). Approx. 80% of dust extracts exhibited significant adipogenic activity at concentrations that are comparable to estimated exposure for children and adults (e.g. ∼20μg/well dust) via either triglyceride accumulation (65%) and/or pre-adipocyte proliferation (50%). Approx. 76% of samples antagonized thyroid receptor beta (TRβ), and 21% activated peroxisome proliferator activated receptor gamma (PPARγ). Triglyceride accumulation was significantly correlated with TRβ antagonism. Sixty-five contaminants were detected in at least 75% of samples; of these, 26 were correlated with adipogenic activity and ten with TRβ antagonism. Small-scale vinifications were performed and wines were characterized. Agronomic management system affected the fungal community composition Hanseniaspora, Areobasidium and Botrytis genera represented 30%, 20% and 10% of the total reads in all samples. A. pullulans, and Cladosprium cladosporioides mainly occurred on organic and biodynamic grapes. Saccharomyces and Pseudopithomyces genera were present only on organic or biodynamic grapes, resp. The agronomic managements also influenced the potential functionality of microbial community. In fact, the metabolic function was increased in organic and biodynamic grapes. Polymers were used only by organic and biodynamic microbial communities, which also showed the highest values of Shannon ′s diversity index, and substrate richness. However, no significative differences were observed for the oenol. parameters analyzed, with the only exception of the content of sugars which were higher on conventional grapes. The fermentation profiles showed that conventional wines had a lower residual sugars content, and a higher amount of alc. The differences observed in the volatile composition of the wines were both quant. and qual. Conventional wines showed a lower content of esters, and a higher concentration of alcs. than organic and biodynamic ones. Biodynamic wines were characterized by the highest content of organic acids. The obtained results revealed that farming practices shape the fungal community influencing wine traits linking the wine with the viti-vinicultural area of origin.
