The present study investigates the influence of pressure on the products of fast pyrolysis of a lignocellulosic biomass (Walnut Shells). Experiments were carried out using a special heated strip reactor (HSR) at temperature of 1573 and 2073 K, pressure up to 8 bar and heating rate of 104 K/s. Two atmospheres have been investigated: N2 and CO2. Increasing the pressure of heat treatment, the combustion pattern and the average combustion reactivity of the chars change remarkably. Multiple components with different combustion reactivity have been found in the char samples. Upon heat treatment in N2 the most reactive components are depleted more easily than upon heat treatment in CO2. Consequently, thermodeactivation is more severe in N2 than in CO2. Raman anal. of the chars shows that graphitic order develops only upon heat treatment at 2073 K at the pressure of 2 bar, but not upon heat treatment at 1573 K, not even at the higher pressure investigated (4 and 8 bar). The loss of reactivity induced by pressure is therefore ascribed to changes in the internal distribution of char components. The evolution of the solid components is accompanied by changes in the distribution of aliphatics/methoxy aromatic compounds in the produced tar. The interplay of temperature, pressure, and CO2 affects both mineral matter and the carbon matrix resulting into a rather complex behavior. The composite PCMs can be processed into a variety of shapes by using molds. The paraffin/NPGDMA/BN composite PCM exhibited a significantly improved shape stability and large phase change enthalpy of 123.0 J/g. Adipocyte fatty acid-binding protein (A-FABP, also called FABP4, aP2) is an adipokine identified as a critical regulator of metabolic function due to its dual functions of fatty acid transport and pro-inflammation. Because of the high therapeutic potential of A-FABP inhibition for the treatment of metabolic diseases and related vascular complications, numerous inhibitors have been developed against A-FABP. However, none of these inhibitors have been approved for use in patients due to severe side effects. Here, we used a virtual screening (VS) strategy to identify potential inhibitors of A-FABP in the latest FDA-approved drug library (∼2600 compounds), aiming to explore the existing drugs with proven safety profiles. We firstly combined ligand-based machine learning and structure-based mol. docking to develop a screening pipeline for identifying A-FABP inhibitors. The screening of FDA-approved drugs identified four compounds (Cobimetinib, Larotrectinib, Pantoprazole, and Vildagliptin) with the highest scores, whose inhibitory effects on A-FABP were further assessed in cellular assays. Among the selected compounds, Cobimetinib significantly inhibited the activation of the JNK/c-Jun signaling pathway by A-FABP in mouse macrophages without causing obvious cytotoxicity. In summary, we present an integrated VS pipeline for A-FABP inhibitor screening, and identified Cobimetinib as a novel A-FABP inhibitor that may be repurposed for the treatment of metabolic diseases and associated vascular complications. Their total concentrations ranged from 38.9 to 3810 μg/g (median of 193 μg/g) and from 18.6 to 1910 μg/g (307 μg/g) in lipstick (n = 34) and nail polish (n = 15), resp. The chem. compositions differed between lipstick and nail polish, but non-PAE plasticizers generally dominated over other groups of chems. A number of emerging plasticizers, OPEs, and UV stabilizers have rarely been reported in personal care products or environmental samples but were found at very high levels in beauty products, raising concern about their environmental release and human exposure risk. Research suggests that thyroid cancer incidence rates are increasing, and environmental exposures have been postulated to be playing a role. To explore this possibility, we conducted a pilot study to investigate the thyroid disrupting bioactivity of chem. mixtures isolated from personal silicone wristband samplers within a thyroid cancer cohort. Specifically, we evaluated TRβ antagonism of chem. mixtures extracted from wristbands (n = 72) worn by adults in central North Carolina participating in a case-control study on papillary thyroid cancer. Sections of wristbands were solvent-extracted and analyzed via mass spectrometry to quantify a suite of semivolatile chems. A second extract from each wristband was used in a bioassay to quantify TRβ antagonism in human embryonic kidney cells (HEK293/17) at concentrations ranging from 0.1 to 10% of the original extract (by volume). Approx. 70% of the sample extracts tested at a 1% extract concentration exhibited significant TRβ antagonism, with a mean of 30% and a range of 0-100%. Inhibited cell viability was noted in >20% of samples that were tested at 5 and 10% concentrations Antagonism was pos. associated with wristband concentrations of several phthalates, organophosphate esters, and brominated flame retardants. These results suggest that personal passive samplers may be useful in evaluating the bioactivities of mixtures that people contact on a daily basis. We also report tentative associations between thyroid receptor antagonism, chem. concentrations, and papillary thyroid cancer case status.
