Abstract: Background: The most frequently diagnosed primary brain tumor is glioblastoma (GBM). Nearly all patients experience tumor recurrence and up to 90% of which is local recurrence. Thus, increasing the therapeutic ratio of radiotherapy using hypofractionated stereotactic radiotherapy (HSRT) can reduce treatment time and may increase tumor control and improve survival. To evaluate the efficacy and toxicity of the combination of HSRT and intensity-modulated radiotherapy (IMRT) with temozolomide after surgery in GBM patients and provide evidence for further randomized controlled trials. Methods/design: HSCK-010 is an open-label, single-arm phase II trial (NCT04547621) which includes newly diagnosed GBM patients who underwent gross total resection. Patients will receive the combination of 30 Gy/5fx HSRT, and 20 Gy/10fx IMRT adjuvant therapy with concurrent temozolomide and adjuvant chemotherapy. The primary endpoint is overall survival (OS). Secondary outcomes include progression-free survival (PFS) rate, objective-response rate (ORR), quality of life (Qol) before and after the treatment, cognitive function before and after the treatment, and rate of treatment-related adverse events (AE). The combination of HSRT and IMRT with temozolomide can benefit the patients after surgery with good survival, acceptable toxicity, and reduced treatment time. Trial registration: NCT04547621. Registered on 14 Sept. 2020.

Jian-Gan-Xiao-Zhi decoction (JGXZ) is a traditional Chinese medicine formula to treat patients with non-alc. fatty liver disease (NAFLD). The study aimed to analyze the mechanism of JGXZ in adipocytes and detect the main components of the drug in rat serum. 3T3-L1 preadipocytes were used to establish an insulin resistant (IR) adipocyte model. Lipid accumulation in adipocytes was detected by oil red O staining. After JGXZ treatment, glucose consumption, total cholesterol (TC), and triglyceride (TG) were analyzed using the corresponding kits. ROS levels were measured by flow cytometry. In addition, Western blot was used to assess LKB1/AMPK and JNK/IRS/PI3k/AKT expressions. The main components of JGXZ in rat serum samples were detected by LC-MS/MS using a Phenomenex Luna C18 column, a mobile phase of methanol and 0.1% formic acid solution, and ESI detection. JGXZ significantly decreased glucose levels and adipogenesis, accompanied by decreased IR (P < 0.01). Besides, JGXZ markedly affected ROS, LKB1/AMPK, and JNK/IRS/PI3k/AKT levels (P < 0.01). R1, Rg1, paeoniflorin, Rb1, astragaloside IV, and tanshinone could be significantly quantified. JGXZ decreased glucose and lipid synthesis, possibly via the ROS/AMPK/JNK pathway. R1, Rg1, paeoniflorin, Rb1, astragaloside IV, and tanshinone in JGXZ could play major roles in treating NAFLD, which could assist in the study of the mechanism of JGXZ in treating NAFLD. Eighteen compounds were identified from methanol extract of Pterocarya tonkinensis, including two new natural products 1 and 2: 1, pterocaryalactone; 2, pterocaryafuranone; 3, (1S, 6R)-9-hydroxymegastigm-7-en-3-one; 4, (S)-di-Me malate; 5, α-linolenic acid; 6-8, α-tetralones; 9, (R)-methyl-2-hydroxyl-3-phenyl-propanoate; 10, (E)-4-hydroxycinnamic acid Me ester; 11-14, diarylheptanoids, and 15-18, pentacyclic triterpenoids. Based on results of in vitro antifungal assay, Magnaporthe oryzae and Phytophthora infestans were most sensitive to isolated compounds among tested phytopathogenic fungi. When ten active compounds were applied onto plants at concentration of 100 or 500 μg mL-1, compounds 1 and 8 effectively suppressed rice blast disease, and compounds 15 and 16 not only strongly reduced development of blast on rice but also effectively controlled the development of late blight on tomatoes. This is first report to evaluate the in vitro and in vivo antifungal activities of the isolated compounds from a methanol extract of Pterocarya tonkinensis against phytopathogenic fungi, and our results suggest that Pterocarya tonkinensis and its substances can be used as source to develop natural fungicides.

In this paper, the waste polyethylene terephthalate (PET) was glycolyzed by trimethylolpropane with zinc acetate as catalyst. The effects of different content glycolysis product of waste PET on the appearance, viscosity, particle size and mol. weight of autoxidative water-reducible alkyd resins and the corresponding film adhesion, flexibility, impact resistance, gloss, hardness and chem. resistance were studied. Meanwhile, exptl. results were compared with com. water-reducible alkyd and water-reducible alkyd without the glycolysis product of waste PET. The results show that the maximum concentration of PET in autoxidative waterreducible alkyd resins can reach 8.5 weight%, and the mol. weight, particle size and viscosity of water-reducible alkyd resin do not change much with the increase of PET concentration The introduction of PET resulted in the viscosity of waterreducible alkyd resins being greater than that of water-reducible alkyd resin without PET; this is mainly because PET contains harder terephthalic acid monomer units. However, the particle size of water-reducible alkyd resins with waste PET is significantly lower than that of the water-reducible alkyd resin without PET; this is due to PET-free water-reducible alkyd resin containing more pentaerythritol with greater steric hindrance. In addition, the hardness of the water-reducible alkyd resin paint film (PET content is 8.5%) reaches 1H, which is higher than the hardness (HB) of the water-reducible alkyd resin paint film without PET and the com. alkyd resin paint film, while the phys. properties and chem. resistance of the former are comparable to those of the latter two kinds of paint films. Therefore, the use of waste PET in water-borne coatings systems not only reduces the cost of coatings, but also opens up a new market for recycled PET, which may contribute a promising method for management of waste PET.
