Rebane, Riin;Herodes, Koit research published 《 Matrix interference in LC-ESI-MS/MS analysis of metanephrines in protein precipitated plasma samples》, the research content is summarized as follows. Metanephrine and normetanephrine are measured in blood plasma to diagnose different diseases. Simpler sample preparation procedures are preferred but tend to yield less purified extracts Therefore, thorough investigation of matrix effects is required. In this work, several sample preparation methods and chromatog. modes were compared for liquid chromatog. tandem mass spectrometric (with electrospray ionization; LC-ESI-MS/MS) anal. of metanephrine and normetanephrine in blood plasma. Protein precipitation with methanol was found to be sufficient for sample preparation and pentafluorophenyl column provided adequate chromatog. separation A new cheaper and less labor-intensive approach is proposed where necessary quantitation limits are achieved through a sample preparation containing only protein precipitation and dilution of the sample extract Matrix effects for different sample preparation methods and the use of isotope-labeled internal standards were evaluated. Unusual interference to D3-labeled internal standard of normetanephrine was discovered – signal of interfering compound increased while the matrix effects were reduced by dilution, e.g. dilution eliminates matrix suppression on interfering compound The results stress the need to monitor interfering compounds and evaluate matrix effects at every step of method development. Matrix effects and interferences can be different for analytes and their corresponding isotopically labeled internal standards This means that the use of isotopically labeled internal standards cannot guarantee accuracy of obtained results. New method allows quantification of the low nanomolar concentrations of metanephrine and normetanephrine in plasma samples.

Ravi, Srimadhavi;Barui, Sugata;Kirubakaran, Sivapriya;Duhan, Parul;Bhowmik, Kaushik research published 《 Synthesis and Characterization of Quinoline-3-Carboxamide Derivatives as Inhibitors of the ATM Kinase》, the research content is summarized as follows. Background: The importance of inhibiting the kinases of the DDR pathway for radiosensitizing cancer cells is well established. Cancer cells exploit these kinases for their survival, which leads to the development of resistance towards DNA damaging therapeutics. Objective: In this article, the focus is on targeting the key mediator of the DDR pathway, the ATM kinase. A new set of quinoline-3-carboxamides, as potential inhibitors of ATM, is reported. Methods: Quinoline-3-carboxamide derivatives were synthesized and cytotoxicity assay was performed to analyze the effect of mols. on different cancer cell lines like HCT116, MDA-MB-468, and MDA-MB-231. Results: Three of the synthesized compounds showed promising cytotoxicity towards a selected set of cancer cell lines. Western Blot anal. was also performed by pre-treating the cells with quercetin, a known ATM upregulator, by causing DNA double-strand breaks. SAR studies suggested the importance of the electron-donating nature of the R group for the mol. to be toxic. Finally, Western-Blot anal. confirmed the down-regulation of ATM in the cells. Addnl., the PTEN neg. cell line, MDA-MB-468, was more sensitive towards the compounds in comparison with the PTEN pos. cell line, MDA-MB-231. Cytotoxicity studies against 293T cells showed that the compounds were at least three times less toxic when compared with HCT116. Conclusion: In conclusion, these experiments will lay the groundwork for the evolution of potent and selective ATM inhibitors for the radio- and chemo-sensitization of cancer cells.

Rashed, Nurnobi Md.;Masuda, Koichiro;Ichitsuka, Tomohiro;Koumura, Nagatoshi;Sato, Kazuhiko;Kobayashi, Shu research published 《 Zirconium Oxide-Catalyzed Direct Amidation of Unactivated Esters under Continuous-Flow Conditions》, the research content is summarized as follows. A sustainable and environmentally benign direct amidation reaction of unactivated esters with amines was developed in a continuous-flow system. A com. available amorphous zirconium oxide was found to be an efficient catalyst for this reaction. While the typical amidation of esters with amines required a stoichiometric amount of a promoter or metal activator, the present continuous-flow method enabled the direct amidation reaction under additive-free conditions with an extensive diversity towards various functional groups. High yields of the products were obtained with a nearly equimolar proportion of starting materials to reduce byproduct formation, which rendered this process applicable for use in a sequential-flow system.

Polgar, Alexander M.;Huang, Shine H.;Hudson, Zachary M. research published 《 Donor modification of thermally activated delayed fluorescence photosensitizers for organocatalyzed atom transfer radical polymerization》, the research content is summarized as follows. Thermally activated delayed fluorescence (TADF) photosensitizers based on 9,10-dihydro-9,9-dimethylacridine/2,4,6-triphenylpyrimidine conjugates with strong visible absorption, large excited state reduction potentials, and long-lived triplet excited states were successfully employed in the organocatalyzed atom transfer radical polymerization (O-ATRP) of methacrylic monomers. A donor-modification strategy dramatically improved the stability of the photocatalyst radical cations, while retaining their high oxidizing strengths, a key requirement for controlled O-ATRP. Time-resolved photoluminescence studies of the catalysts support initiation by electron transfer from both singlet and triplet states, with functionalized donors producing higher driving forces for photoinduced electron transfer. A donor-modified TADF photocatalyst was found for the synthesis of methacrylic polymers with D below 1.3 at catalyst loadings down to 50 ppm. This catalyst was also successfully applied in block copolymer synthesis, while the unfunctionalized analog yields entirely uncontrolled polymerization

Poirier, Marion;Pujol-Gimenez, Jonai;Manatschal, Cristina;Buhlmann, Sven;Embaby, Ahmed;Javor, Sacha;Hediger, Matthias A.;Reymond, Jean-Louis research published 《 Pyrazolyl-pyrimidones inhibit the function of human solute carrier protein SLC11A2 (hDMT1) by metal chelation》, the research content is summarized as follows. Solute carrier proteins (SLCs) control fluxes of ions and mols. across biol. membranes and represent an emerging class of drug targets. SLC11A2 (hDMT1) mediates intestinal iron uptake and its inhibition might be used to treat iron overload diseases such as hereditary hemochromatosis. Here we report a micromolar (IC50 = 1.1 μM) pyrazolyl-pyrimidone inhibitor of radiolabeled iron uptake in hDMT1 overexpressing HEK293 cells acting by a non-competitive mechanism, which however does not affect the electrophysiol. properties of the transporter. Isothermal titration calorimetry, competition with calcein, induced precipitation of radioactive iron and cross inhibition of the unrelated iron transporter SLC39A8 (hZIP8) indicate that inhibition is mediated by metal chelation. Mapping the chem. space of thousands of pyrazolo-pyrimidones and similar 2,2′-diazabiaryls in ChEMBL suggests that their reported activities might partly reflect metal chelation. Such metal chelating groups are not listed in pan-assay interference compounds (PAINS) but should be checked when addressing SLCs.

Plamondon, Samuel J.;Warnica, Josephine M.;Kaldre, Dainis;Gleason, James L. research published 《 Hydrazide-Catalyzed Polyene Cyclization: Asymmetric Organocatalytic Synthesis of cis-Decalins》, the research content is summarized as follows. Polyene cyclizations offer rapid entry into terpenoid ring systems. Although enantioselective cyclizations of (E)-polyenes to form trans-decalin ring systems are well precedented, highly enantioselective cyclizations of (Z)-polyenes to form the corresponding cis-decalins have not been reported. Here, we describe the first application of iminium catalysis to the initiation of polyene cyclizations. Et 1,2-diazepane-1-carboxylate catalyzes the cyclization of polyenes bearing enal initiators. Moreover, chiral bicyclic hydrazides catalyze the cyclizations of (Z)-polyene substrates to form cis-decalins with enantioselectivities of up to 97:3 er. DFT calculations suggest the catalysts promote the reaction by stabilizing pos. charge as it develops during the bicyclization.

The bidentate ligand facilitates the nucleophilic addition of the aryl halides RC(=CH2)C(O)Cl (R = Me, hexyl, Bn) to the amide carbonyls R1C(O)NHR2 (R1 = 2-bromophenyl, 2-bromo-4,5-difluorophenyl, 2-bromothiophene-3-yl, 3-bromo-1-methyl-1H-indole-2-yl, etc.; R2 = Me, i-Pr, t-Bu, cyclopropyl, Ph), followed by 1,4-acyl transfer and cross-coupling to obtain 2-benzazepin-5-ones e.g., I and benzo[c]pyrano[2,3-e]azepines e.g., II. The tridentate ligand promotes the selective 7-endo cyclization/cross-coupling to access to 2-benzazepin-3-ones e.g., III. The protocol operates under mild reaction conditions with divergent cyclization patterns that can be easily modulated through the ligand backbone.

Pihtili, Guzin;Biryan, Fatih research published 《 Electrical investigation on Eu+3– doped copolymer composite system and conventional kinetic analysis》, the research content is summarized as follows. Poly(maleic anhydride-co-benzyl methacrylate) copolymer was prepared by using free radical polymerization (FRP) method at 70°C. The structural characterization of the polymer was accomplished FT-IR, 1H and 13C NMR spectroscopic techniques. P(MA-co-BMA)/ EuCl3 composites were prepared rate of 2%, 5% and 10% (weight/weight). The thermal behavior of pure copolymer was investigated by thermogravimetric anal. (TGA) and differential scanning calorimetry (DSC). Thermal decomposition kinetics of poly(MA-co-BMA) carried out from Flynn-Wall-Ozawa method via TG curves for calculated apparent activation energies. According to FWO; the activation energy (Ea) was estimated as; 185.61 kJ/mol. Dielec. and elec. properties of copolymer/composite systems were investigated against frequency (range from 100 Hz to 20 kHz) at room temperature by using an impedance analyzer. The effect of EuCl3 concentration on dielec. constant, dielec. loss factors, ac conductivity, susceptance (B) and impedance (Z) was investigated. A remarkable increase was observed in dielec. and a.c. elec. properties were observed with the addition of Eu+3 to the polymer matrix. The B and Z parameters of poly(MA-co-BMA) and composites were investigated depending on frequency at room temperature based on the a.c. (AC).
