A new approach for balancing the microbial synthesis of ethyl acetate and other volatile metabolites during aerobic bioreactor cultivations

Ethyl acetate is an organic solvent with many industrial applications, currently produced by energy-intensive chemical processes based on fossil carbon resources. Ethyl acetate can be synthesized from renewable sugars by yeasts like Kluyveromyces marxianus in aerobic processes. However, ethyl acetate is highly volatile and thus stripped from aerated cultivation systems which complicate the quantification of the produced ester. Synthesis of volatile metabolites is commonly monitored by repeated analysis of metabolite concentrations in both the gas and liquid phase. In this study, a model-based method for quantifying the synthesis and degradation of volatile metabolites was developed. This quantification of volatiles is solely based on repeatedly measured gas-phase concentrations and allows calculation of reaction rates and yields in high temporal resolution. Parameters required for these calculations were determined in abiotic stripping tests. The developed method was validated for ethyl acetate, ethanol and acetaldehyde which were synthesized by K. marxianus DSM 5422 during an iron-limited batch cultivation; it was shown that the presented method is more precise and less time-consuming than the conventional method. The biomass-specific synthesis rate and the yield of ethyl acetate varied over time and exhibited distinct momentary maxima of 0.50 g g(-1)h(-1) and 0.38 g g(-1) at moderate iron limitation.

The transfection efficiency of siRNA mediated by cationic polymers is limited due to the instability of polymers/siRNA complexes in the presence of serum. Poly(ethylene glycol) (PEG) is usually applied to modify cationic polymers, so as to reduce protein and cell adsorption and then to improve siRNA transfection efficiency. However, the polymers’ modification with PEG mostly consumes the free amino of the polymers, which can, in turn, reduce the charge density and limit their siRNA transfection efficacy. Here, a new PEG modification strategy that need not consume the surface aminos of polymers is proposed. Catechol-PEG polymers are coated on the surface of phenylboronic acid (PBA)-modified Generation 5 (G5) poly(amidoamine) dendrimers (G5PBA) via reversible boronate esters to establish PEG-modified dendrimer/siRNA nanoassemblies for efficient siRNA delivery. The PEG/G5PBA/siRNA nanoassemblies have positive charge and show excellent gene silencing efficacy in the absence of serum in vitro. More importantly, the PEG/G5PBA/siRNA nanoassemblies also exhibit excellent serum resistance and gene silencing efficacy in serum-containing medium. Furthermore, the effective antiserum and gene silencing efficacy elicited by these nanoassemblies lead to excellent antitumor effects in vivo. Columnar self-assembly, electrochemical and luminescence properties of basket-shaped liquid crystalline derivatives of Schiff-base-moulded p-tert-butyl-calix[4]arene

It was observed that all the prepared target compounds exhibited enantiotropic mesophases with good thermal stability. The structural and conformational characterisation of these new compounds was achieved using various spectroscopic techniques, including FT-IR, H-1 NMR, C-13 NMR and MALDI-TOF. The relationship between the structure and the mesomorphic behaviour of the compounds containing an ethyl bridge with linked ester groups (5a-5d) is discussed. This new class of supramolecular derivatives is promising, especially considering the emissive nature and stabilisation of their columnar hexagonal mesophases, and their high range of thermal stability.

Aliphatic poly(ester-carbonates) are receiving extensive research attention as tailorable materials suitable for multiple applications from tissue engineering and 3D scaffold printing to drug delivery. In this work, we report on one-pot synthesis of random copolymers of TMC and delta-VL using metal-free biocompatible 1,5,7-triazabicyclo[4.4.0]dec-5-ene as a catalyst and benzyl alcohol and poly(ethylene oxide) as initiators. Random poly(ester-carbonates) with TMC : VL unit ratios ranging from 80 : 20 to 20 : 80 were synthesized via ring-opening polymerization while displaying excellent agreement of comonomers’ ratios in the feed and copolymer chains. The copolymers’ supramolecular structure, thermal and mechanical properties were thoroughly analyzed by various methods. The obtained results clearly indicated that the physicochemical properties can be controlled simply by varying the ratio of comonomers and the length of segments in the copolymer chain. Several copolymers exhibited behavior of thermoplastic elastomers with the most promising one exhibiting a 2200% increase in elongation at break compared to the poly(valerolactone) homopolymer while retaining tensile strength and Young’s modulus suitable for biomedical applications. Overall, our work contributed to widening the portfolio of tailorable copolymers for specialized bioapplications and possibly paving a way for the use of more sustainable polymers in the biomedical field.

We found that compounds 1-4 exhibited potent anti-inflammatory effects, as indicated by suppression of lipopolysaccharide-stimulated nitric oxide and cytokine production with 50% inhibitory concentrations between 20.09 mu M and 27.63 mu M. These results identify two novel quinones from J. mandshurica with potential utility as anti-inflammatory compounds.
