Antimyeloma Potential of Caffeic Acid Phenethyl Ester and Its Analogues through Sp1 Mediated Downregulation of IKZF1-IRF4-MYC Axis

Caffeic acid phenethyl ester (CAPE, 2), a natural compound from propolis, is a well-documented antitumor agent with nuclear factor kappa B (NF-.B) inhibitory activity. Key transcription factors regulated by NF-kappa B, namely, interferon regulatory factor-4 (IRF4) and octameric binding protein-2 (OCT2), are implicated in the tumorigenesis of multiple myeloma (MM), an incurable bone marrow cancer. Adverse effects and resistance to current chemotherapeutics pose a great challenge for MM treatment. Hence, the structure-activity relationships of CAPE (2) and 21 of its analogues were evaluated for their antimyeloma potential. Preclinical evaluation revealed that CAPE (2) and the 3-phenylpropyl (4), 2,5-dihydroxycinnamic acid 3-phenylpropyl ester (17), and 3,4-dihydroxycinnamic ether (22) analogues inhibited human myeloma cell growth. Analogue 4 surpassed CAPE (2) and lenalidomide in showing strong apoptotic effects with a remarkable decrease in IRF4 levels. The analogue 17 exhibited the most potent anti-MM activity. The downregulation of specificity protein 1 (Sp1) and the IKZF1-IRF4-MYC axis by CAPE (2) analogues 4 and 17 revealed their novel mechanism of action. The analogues showed no adverse cytotoxic effects on normal human cells and exhibited appropriate in silico pharmacokinetic properties and drug-likeness. These findings suggest the promising application of CAPE (2) analogues to target Ikaros (IKZF1)/IRF4 addiction, the so-called Achilles heel of myeloma, for better treatment outcomes.

Self-Assembly of a Multi-Functional Hydrogel from a Branched Peptide Amphiphile and Its Effects on Bone Marrow Mesenchymal Stem Cells

In this work, a branched peptide amphiphile (B-PA) presenting RGD and IKVAV motifs was fabricated by solid-phase peptide synthesis and self-assembled into a nanofiber hydrogel in which rabbit bone marrow mesenchymal stem cells (BMSCs) were seeded and cultured for seven days. Specifically, 1 wt% B-PA was self-assembled into a nanofiber hydrogel with the addition of culture medium and observed using transmission electron microscopy. The B-PA with a molecular weight of 2191.72 and a purity >95% self-assembled into nanofibers with diameters from 6 to 8 nm and lengths ranging from hundreds of nanometers to several micrometers. BMSCs were acquired from rabbits using differential adherence methods and identified by flow cytometry for cell phenotype. The cells were stained with calcein acetoxymethyl ester/propidium iodide to assess cell viability, CCK-8 to assess cell cytotoxicity and proliferation, and Hochest 33342 to assess cell adhesion. They were also immunofluorescently labeled with microtubule-associated protein-2 (MAP-2), neurofilament protein (NF), and glial fibrillary acidic protein (GFAP) to assess cell transdifferentiation. The B-PA hydrogel provided platform upon which the CD29+/44+ cells adhered and proliferated, and it induced the transdifferentlation of cells into neural cells expressing the markers MAP-2, NF, and GEAR The hydrogel exhibited good cytocompatibility and multiple functions, and may therefore serve as a scaffold for neural tissue engineering.

Study on Hydrolysis of Magnesium Hydride by Interface Control

Magnesium hydride (MgH2) is one of the competitive hydrogen storage materials on account of abundant reserves and high hydrogen content. The hydrolysis of MgH2 is an ideal and controllable chemical hydrogen generation process. However, the hydrolyzed product of MgH2 is a passivation layer on the surface of the magnesium hydride, which will make the reaction continuity worse and reduce the rate of hydrogen release. In this work, hydrogen generation is controllably achieved by regulating the change of the surface tension value in the hydrolysis, a variety of surfactants were systematically investigated for the effect of the hydrolysis of MgH2 In the meantime, the passivation layer of MgH2 was observed by scanning electron microscope (SEM), and the surface tension value of the solution with different surfactants were monitored, investing the mechanism of hydrolysis adding different surfactants. Results show that different surfactants have different effects on hydrogen generation. The hydrogen generation capacity from high to low is as follows: tetrapropylammonium bromide (TPABr), sodium dodecyl benzene sulfonate (SDBS), Ecosol 507, octadecyl trimethyl ammonium chloride (OTAC), sodium alcohol ether sulfate (AES), and fatty methyl ester sulfonate (FMES-70). When the ratio of MgH2 to TPABr was 5 : 1, the hydrogen generation was increased by 52% and 28.3%, respectively, at the time of 100 s and 300 s. When hydrolysis time exceeds 80 s, the hydrogen generation with AES and FMES-70 began to decrease; it was reduced by more than 20% at the time of 300 s. SEM reveals that surfactants can affect the crystalline arrangement of Mg(OH)(2) and make the passivation layer three-dimensionally layered providing channels for H2O molecules to react with MgH2.

Preparation of Montelukast Sodium and Graphene Nanomaterials for the Treatment of Asthma

This study focuses on the biomaterial effects of montelukast sodium and graphene oxide (GO) on the onset of asthma disease. First, the synthesis process was adopted, with 2-[3-(S)-[3-[2-(7-chloro-2-quinolinyl) vinyl] phenyl]-3-hydroxypropyl] benzyl ester as raw materials, to synthesize montelukast sodium, and then Hummers method was used to obtain graphene oxide (GO), and GO was reduced to obtain graphene (EG). After the preparation completed, the purity of montelukast sodium was tested by liquid chromatography (HPLC), and the structural analysis of graphene nanomaterials was performed by X-ray diffractometer and Raman spectroscopy. Firstly, 30 mice were selected to observe the effect of montelukast sodium on the number of Th17 and cytokine IL-17 levels in asthmatic mice, then 50 mice were selected to observe whether the graphene-based nanomaterials had little effect of oxidative stress index in rat lung tissue. In the experiment, the spectrum analysis and liquid chromatography analysis showed that the purity of the prepared montelukast sodium exceeded 99%. The prepared graphene nanomaterials showed a strong D peak at 1037 cm(-1) by Raman spectroscopy.
